Phosphate, calcium, and vitamin D signaling, transport, and metabolism in the endometria of cyclic ewes

Claire Stenhouse et al.

Summary: Recent evidence shows the important roles of progesterone and interferon tau in the regulation of calcium, phosphate, and vitamin D signaling in the uteri of pregnant sheep. However, the effects of progesterone and estradiol, and their respective receptors, on mineral signaling during the estrous cycle have not been investigated. This study found that the expression of certain genes and proteins related to mineral signaling was different at different stages of the estrous cycle. Overall, these results suggest that progesterone and its receptor play important roles in regulating mineral signaling during the estrous cycle in ewes.

USP18 promotes endometrial receptivity via the JAK/STAT1 and the ISGylation pathway

Xinyan Zhang et al.

Summary: In this study, the role and mechanism of Ubiquitin-specific protease 18 (USP18) on endometrial receptivity in goats were investigated. The results showed that USP18 promotes endometrial receptivity by regulating the JAK/STAT1 pathway and ISGylation. Additionally, the expression levels of USP18 were significantly increased during early pregnancy. Therefore, USP18 may play an important role in goat endometrial receptivity.

Effects of novel variants in BMP15 gene on litter size in Mongolia and Ujimqin sheep breeds

Yilin Wang et al.

Summary: Reproductive traits are important in the sheep industry, and the BMP15 gene plays a major role. This study identified 12 novel variants in the BMP15 gene of Mongolia sheep, and found significant associations between certain variants and litter size in Mongolia ewes and Ujimqin ewes. One of the mutations is predicted to affect the tertiary structure of BMP15. These findings may contribute to improving litter size in sheep.

Towards a Better Understanding of Endometriosis-Related Infertility: A Review on How Endometriosis Affects Endometrial Receptivity

Jing Shan et al.

Summary: Endometriosis is the leading cause of infertility, and its mechanisms leading to implantation failure include hormone imbalance, inflammation, and immune regulatory dysfunction. This review provides an overview of current research on endometriosis-related infertility/subfertility, focusing on factors such as genetic, epigenetic, glycosylation, metabolism, and microRNA. The future clinical implications of these factors in the diagnosis and therapy to improve endometrial function are also discussed.

Qianying Yang et al.

Summary: This study provides the first comprehensive proteomic map of ligand-receptor pathway cascades at the maternal-fetal interface in sheep. It reveals the essential role of these cascades in cell adhesion and invasion, redox homeostasis, and the immune response. The study also demonstrates the physical interaction of albumin and claudin 4 and their roles in embryo attachment to the endometrium, as well as the novel function of enhanced conceptus glycolysis in remodeling uterine receptivity by inducing endometrial histone lactylation.

BMP/SMAD1/5 Signaling in the Endometrial Epithelium Is Essential for Receptivity and Early Pregnancy

Suni Tang et al.

Summary: The biological processes that control endometrial receptivity and embryo implantation are critical for the successful outcome of pregnancy. Researchers found that the uterine epithelial BMP/SMAD1/5 signaling pathway plays a key role during early pregnancy and its deletion may lead to defects in endometrial receptivity and embryo implantation issues.

Global proteomic analysis of preimplantational ovine embryos produced in vitro

Jose Renato S. Passos et al.

Summary: The present study characterized the major proteome of preimplantation ovine embryos grown in vitro. The analysis identified proteins that are involved in translation, protein transport, and stabilization. Functional cluster analysis revealed differences between the in vitro and in vivo proteomes of sheep embryos, indicating that in vitro derived blastocysts have distinct protein profiles compared to those produced in vivo.

Melatonin improves pregnancy outcomes in adenomyosis mice by restoring endometrial receptivity via NF-kappa B/apoptosis signaling

Xiaohong Guan et al.

Summary: In this study, the effect of melatonin on uterine receptivity and pregnancy outcomes in an adenomyosis mouse model was investigated. The results showed that melatonin significantly improved implantation rates and restored impaired uterine development and endometrial receptivity in the adenomyosis mice. Melatonin also mitigated adenomyosis-induced changes in the uterine microenvironment and improved pregnancy outcomes by inhibiting the NF-kappa B signaling pathway and decreasing endometrial cell apoptosis. These findings provide new insight into melatonin-based therapy for adenomyosis-related infertility.

Main actors behind the endometrial receptivity and successful implantation

Laura Governini et al.

Summary: Embryo implantation occurs during a specific period in the mid-secretory phase of the menstrual cycle, and interaction between the endometrium and blastocyst is crucial for successful implantation. While no single molecule or receptor has been identified as essential for implantation, a variety of mediators support pregnancy establishment. Understanding these complex mechanisms can help address issues of implantation failure and infertility.
