Cancer statistics, 2023

Rebecca L. Siegel et al.

Summary: The American Cancer Society predicts that there will be 1,958,310 new cancer cases and 609,820 cancer deaths in the United States in 2023. While incidence trends for most cancers are favorable, prostate cancer saw a significant increase of 3% annually from 2014 to 2019. However, the overall cancer death rate continues to decline due to advances in treatment, with a 33% reduction since 1991.

CHRNA5 Contributes to Hepatocellular Carcinoma Progression by Regulating YAP Activity

Yan Fu et al.

Summary: In this study, the role of CHRNA5 in hepatocellular carcinoma (HCC) was investigated. Increased expression of CHRNA5 in HCC tissues was found to be positively correlated with tumor stage and worse survival in patients. Both in vitro and in vivo assays showed that CHRNA5 regulates HCC proliferation through the YAP pathway and promotes stemness through stemness-associated genes. Furthermore, CHRNA5 enhances HCC metastasis by regulating epithelial-mesenchymal transition (EMT)-associated genes. It was also observed that CHRNA5 affects the sensitivity of HCC to sorafenib treatment. These findings indicate that CHRNA5 plays a crucial role in HCC progression and drug resistance.

Divergent Roles of α5 and β4 Nicotinic Receptor Subunits in Food Reward and Nicotine-induced Weight Loss in Male Mice

Alberte Wollesen Breum et al.

Summary: The physiological and molecular mechanisms by which nicotine affects energy homeostasis and food-motivated behaviors are not well understood. This study demonstrates the divergent roles of alpha 5 and beta 4 subunits of nicotinic acetylcholine receptors (nAChRs) in food reward and the essential role of beta 4-containing nAChRs in the weight-lowering effects of nicotine in obese mice. The study also suggests a crosstalk between incretin biology and nAChR signaling through beta 4-containing nAChRs. Further research into cholinergic neurotransmission and targeted treatments involving beta 4-containing nAChRs is encouraged.

Small Molecular Inhibitors Block TRPM4 Currents in Prostate Cancer Cells, with Limited Impact on Cancer Hallmark Functions

Anna Borgstroem et al.

Summary: TRPM4 is a widely expressed ion channel that plays a role in cancer hallmark functions, such as viability, migration, cell cycle shift, and adhesion, in prostate cancer cells. However, blocking TRPM4 ion conductivity is not sufficient to prevent its role in these functions.

Nicotine promotes breast cancer metastasis by stimulating N2 neutrophils and generating pre-metastatic niche in lung

Abhishek Tyagi et al.

Summary: Smoking has a profound impact on tumor immunity and tumor progression, with chronic nicotine exposure promoting the formation of a pre-metastatic niche in the lungs by recruiting pro-tumor N2-neutrophils that release lipocalin-2. Salidroside may be used to enhance the anti-tumor activity of neutrophils in breast cancer patients.

CHRNA5 belongs to the secondary estrogen signaling network exhibiting prognostic significance in breast cancer

Huma Shehwana et al.

Summary: The study found that CHRNA5 expression in breast cancer cells was induced by E2 in a dose- and time-dependent manner, with higher levels in ER- tumors than ER+ tumors. Higher CHRNA5 expression was associated with a worse prognosis in untreated breast cancer patients, as well as with secondary E2 signaling. The study uncoupled primary and secondary target genes, revealing distinct clustering patterns in multiple public datasets.
