Brain Waste Removal System and Sleep: Photobiomodulation as an Innovative Strategy for Night Therapy of Brain Diseases

Oxana Semyachkina-Glushkovskaya et al.

Summary: Emerging evidence suggests that sleeping brain activates the brain waste removal system (BWRS) to remove wastes and toxins from the central nervous system (CNS). Meningeal lymphatic vessels (MLVs) play an important role in the BWRS. Night stimulation of the BWRS is being discussed as a promising strategy for neurorehabilitation medicine.

Sleep-A brain-state serving systems memory consolidation

Svenja Brodt et al.

Summary: The mechanisms of long-term memory consolidation during wakefulness and sleep differ, with neuronal replay being a key mechanism during both states. Sleep-dependent memory transformation is intensified during early development, despite the immaturity of the hippocampus. Sleep consolidation is supported by spontaneous hippocampal replay, which aids in memory formation in the neocortex.

The meningeal lymphatic vessels and the glymphatic system: Potential therapeutic targets in neurological disorders

Gaowei Li et al.

Summary: The recent discovery of meningeal lymphatic vessels and glymphatic pathways challenges the previous belief that the central nervous system lacks a lymphatic system. The close connection between these two systems facilitates waste removal and immune cell trafficking, but dysfunction can lead to neurodegenerative diseases. Noninvasive imaging techniques offer potential for studying the human glymphatic-mLV system and diagnosing neurological disorders.

Thalamo-Cortical and Thalamo-Thalamic Coupling During Sleep and Wakefulness in Rats

Ilya Sysoev et al.

Summary: This study investigated changes in functional connectivity between the thalamus and cortex and between different thalamic nuclei during the process of falling asleep and deepening into slow-wave sleep. The findings revealed that higher order nuclei are more inhibited than first-order nuclei during deep slow-wave sleep. These differences are important for the coordination of sleep-related processes.

Reduced Metabotropic Glutamate Receptor Type 5 Availability in the Epileptogenic Hippocampus: An in vitro Study

Maria Zimmermann et al.

Summary: Abnormalities in the expression of mGluR5 have been observed in the hippocampus of patients with drug-resistant mTLE. The present study demonstrated reduced receptor density in mTLE specimens compared to control hippocampi, independent of age, sex, and K-D. The findings suggest that the decreased mGluR5 availability observed in PET studies may be due to internalization or conformational changes of the receptor.

Expert Opinion: Managing sleep disturbances in people with epilepsy

Lino Nobili et al.

Summary: Poor sleep and daytime sleepiness are common in people with epilepsy, which can be exacerbated by seizures and medications. Standardizing diagnostic pathways for sleep-related issues and implementing psychoeducational approaches are recommended for managing sleep disorders in epilepsy patients. Optimal dosing and timing of medications are crucial for improving nocturnal sleep quality and avoiding daytime sedation.

Sleep Disturbances in Rats With Genetic Pre-disposition to Spike-Wave Epilepsy (WAG/Rij)

Evgenia Sitnikova

Summary: WAG/Rij rats are a valid genetic model of absence epilepsy, exhibiting SWDs and a close relationship between slow-wave EEG activity, sleep spindles, and SWDs. Studies on sleep EEG provide important information about epileptogenic processes related to spike-wave epilepsy.

Partial mGlu5 Negative Allosteric Modulator M-5MPEP Demonstrates Antidepressant-Like Effects on Sleep Without Affecting Cognition or Quantitative EEG

Kimberly M. Holter et al.

Summary: Selective negative allosteric modulators (NAMs) targeting the metabotropic glutamate receptor subtype 5 (mGlu(5)) have shown anxiolytic and antidepressant effects, but concerns remain about potential adverse effects. Partial mGlu(5) NAMs, with submaximal levels of blockade, may have a broader therapeutic index than full mGlu(5) NAMs. Studies comparing M-5MPEP and VU0424238 found differences in their effects on sleep, cognition, and brain function, supporting the development of partial mGlu(5) NAMs.
