The Role of COX-2 and PGE2 in the Regulation of Immunomodulation and Other Functions of Mesenchymal Stromal Cells

Agnieszka Kulesza et al.

Summary: The molecular mechanisms underlying the immunomodulatory properties of MSCs and the role of PGE2 and COX-2 in these properties are not fully understood. This article summarizes previous observations regarding the role of PGE2 and COX-2 in the immunomodulatory properties and other vital functions of MSCs. The most consistent results suggest that MSC-derived PGE2 inhibits the early maturation of dendritic cells, suppresses the proliferation of activated lymphocytes, and stimulates the differentiation of macrophages into M2 phenotype. COX-2/PGE2 also plays an important role in maintaining the basic life functions of MSCs and affects the formation of niches conducive to hematopoiesis and carcinogenesis.

Therapeutic Perspectives for Inflammation and Senescence in Osteoarthritis Using Mesenchymal Stem Cells, Mesenchymal Stem Cell-Derived Extracellular Vesicles and Senolytic Agents

Michael G. G. Rizzo et al.

Summary: Osteoarthritis (OA) is the leading cause of disability among the elderly and is increasingly affecting younger individuals. Recent advancements in understanding the pathology of OA have led to the identification of potential therapeutic approaches targeting specific molecular pathways, such as inflammation and cellular senescence. Stem cell therapies and senolytics show promise in controlling patient symptoms and slowing disease progression.

The JNK-EGR1 signaling axis promotes TNF-α-induced endothelial differentiation of human mesenchymal stem cells via VEGFR2 expression

Euitaek Jung et al.

Summary: In the tumor microenvironment, tumor necrosis factor alpha (TNF-alpha) promotes the endothelial differentiation of mesenchymal stem cells (MSCs) through the activation of the JNK-EGR1 signaling axis. This finding is important for identifying potential therapeutic targets for solid tumor vasculatures.

CD146+Endometrial-Derived Mesenchymal Stem/Stromal Cell Subpopulation Possesses Exosomal Secretomes with Strong Immunomodulatory miRNA Attributes

Clarissa Lenero et al.

Summary: This study discovered that endometrial mesenchymal stem/stromal cells (eMSC) and their CD146+ subpopulation possess exosomal secretomes with potent immunomodulatory miRNA attributes. These exosomes are involved in macrophage polarization, T cell activation, and regulation of inflammatory cytokines. The findings suggest that eMSC EXOs could be used for therapeutic purposes in resolving detrimental aspects of tissue inflammation.

Fibroblast Growth Factor 9 (FGF9) negatively regulates the early stage of chondrogenic differentiation

Xiaoyue Zhang et al.

Summary: FGF9 plays a crucial role in early chondrogenesis, partly through the AKT/GSK-3 β pathway, regulating cartilage matrix formation and gene expression. Inhibition of FGF9 suppresses AKT and GSK-3 β phosphorylation, leading to inhibited chondrocyte differentiation, which can be compensated by the activation of AKT.

Mesenchymal Stem Cell-Derived Extracellular Vesicles for Osteoarthritis Treatment: Extracellular Matrix Protection, Chondrocyte and Osteocyte Physiology, Pain and Inflammation Management

Thu Huyen Nguyen et al.

Summary: Osteoarthritis is a common degenerative disease that can lead to persistent pain and motion restriction, and stem cells, particularly mesenchymal stem cells, have been explored as a potential alternative therapy. Extracellular vesicles containing trophic factors are crucial in MSC-based treatment, and can target various biological processes associated with cartilage degradation. However, outcomes of EV treatment for OA can vary depending on the EV cargo, cell source, and pathological condition.

Cell-associated type I collagen in nondegenerate and degenerate human articular cartilage

Katarzyna Styczynska-Soczka et al.

Summary: Abnormal chondrocytes are present in nondegenerate cartilage, suggesting dedifferentiation and production of a weakened matrix. With degeneration, the percentage of normal chondrocytes decreases while abnormal cells and clusters increase, along with collagen type I labelling. The study also found an increase in collagen type I around chondrocytes with degeneration, indicating fibro-cartilaginous repair tissue progression similar to osteoarthritis.

Summary: This study analyzed individual participant-level data from international observational studies to determine the association of knee osteoarthritis with mortality in the general population. The findings showed that knee osteoarthritis-related pain is significantly associated with reduced time-to-mortality, while osteoarthritis itself is not associated with mortality.
