Cellular Red-Ox system in health and disease: The latest update

Atala Bihari Jena et al.

Summary: Cells develop various defense mechanisms to adapt to reactive oxygen species (ROS) exposure, including antioxidant enzymes like SOD, CAT, and GPX, as well as non-enzymatic defenses such as vitamins and polyphenols. This review article explores the production of ROS during oxidation/reduction processes and the engagement of the antioxidant defense system in scavenging ROS.

Non-Alcoholic Fatty Liver Disease or Type 2 Diabetes Mellitus-The Chicken or the Egg Dilemma

Marcin Kosmalski et al.

Summary: In clinical practice, patients often present both non-alcoholic fatty liver disease (NAFLD) and type 2 diabetes mellitus (T2DM). The etiopathogenesis of NAFLD is linked to insulin resistance (IR) and obesity, while T2DM develops concurrently. Studies have not fully elucidated the mechanisms of their coexistence, making it difficult to determine which disease appears first. However, it is crucial to bring attention to the coexistence of NAFLD and T2DM to prevent their consequences.

Impact of metabolic risk factors on hepatic and cardiac outcomes in patients with alcohol- and non-alcohol- related fatty liver disease

Jihye Lim et al.

Summary: This study evaluated the different effects of metabolic risk factors on alcoholic fatty liver disease (AFLD) and non-alcoholic fatty liver disease (NAFLD), and found that metabolic risk factors had a significant impact on hepatic outcomes but a relatively smaller impact on cardiac outcomes. Patients with NAFLD without metabolic risk factors had a lower risk of cardiac outcomes.

Summary: The study revealed that the CAT C262T variant is associated with overall cancer susceptibility, especially in Caucasians and Asians, with significant correlation in various cancers. Subgroup analyses indicated a significant link with specific types of cancers, while different genotyping methods also showed significant association with cancer risk._Meta-regression and trial sequential analysis further supported the validity of the findings.

Non-alcoholic fatty liver disease (NAFLD): a review of pathophysiology, clinical management and effects of weight loss

Sjaak Pouwels et al.

Summary: Given the increasing prevalence of diabetes and obesity worldwide, the deleterious effects of non-alcoholic fatty liver disease (NAFLD) are becoming a growing challenge for public health. NAFLD is closely associated with metabolic disorders and can range from mild forms to severe forms with potential liver failure. Advanced fibrosis is a major predictor of morbidity and liver-related mortality. Diagnosis of NAFLD is established through liver biopsy and treatment options include weight loss, dietary modification, and addressing underlying metabolic syndrome.

NAFLD: Mechanisms, Treatments, and Biomarkers

Fatiha Nassir

Summary: Nonalcoholic fatty liver disease (NAFLD), also known as metabolic-associated fatty liver disease (MAFLD), is a common liver disease worldwide that is closely linked to the obesity epidemic. Currently, there is no specific pharmacological treatment available for NAFLD due to incomplete understanding of its mechanisms, lack of accurate and inexpensive imaging tools, and inadequate non-invasive biomarkers. NAFLD can lead to metabolic-associated steatohepatitis (NASH), liver fibrosis, and hepatocellular carcinoma.

Ethanol Metabolism in the Liver, the Induction of Oxidant Stress, and the Antioxidant Defense System

Martha Lucinda Contreras-Zentella et al.

Summary: The liver metabolizes ethanol through three enzymatic pathways, with alcohol dehydrogenase considered the most important enzyme. Contradicting experiments suggest that non-alcohol dehydrogenase pathways may be more relevant for ethanol metabolism than previously thought. Ethanol can be metabolized to acetaldehyde mainly through cytochrome P450, which generates reactive oxygen species. Catalase can also participate in ethanol metabolism by reacting with ethanol to produce acetaldehyde. Besides its role as an antioxidant, catalase may have a primary role in xenobiotic metabolism in the liver.

The role of mitochondria dysfunction and hepatic senescence in NAFLD development and progression

Siarhei A. Dabravolski et al.

Summary: Senescence plays a crucial role in metabolic disorders and chronic inflammatory diseases, particularly in the development of NAFLD involving hepatocyte senescence. Mitochondria, as the main energy and ROS producing organelle, are central in accelerated senescence and disease development. The regulation of mitochondrial functions and genetic mutations are discussed as potential diagnostic markers and treatment targets for NAFLD, with advanced age being an independent risk factor.

Summary: This article discusses the interplay between oxidative stress and other pathogenic factors in non-alcoholic fatty liver disease (NAFLD), emphasizing the importance of reactive oxygen species (ROS) in NAFLD progression. It also points out the effectiveness of healthy diet and physical activity in NAFLD treatment, but lack of compliance, and the potential of precision medicine in selecting the best treatment for specific patients.

Heterogeneity of non-alcoholic fatty liver disease: Implications for clinical practice and research activity

Partha Pal et al.

Summary: Non-alcoholic fatty liver disease (NAFLD) is a heterogeneous condition with substantial inter-individual variation influenced by various factors. Failure to address this heterogeneity partly explains the sub-optimal response to current therapies. Leading experts have suggested renaming it to metabolic-associated fatty liver disease (MAFLD) to better reflect current knowledge of heterogeneity.
