Cellular and molecular mechanisms of allergic asthma

Zsolt Komlosi et al.

Summary: Asthma is a chronic disease that affects over 350 million people worldwide and is the most common chronic disease in children. It is influenced by genetic and environmental factors, with allergic eosinophilic asthma being the most common subtype. The prevalence of asthma has increased significantly in industrialized countries in the last 60 years, possibly due to increased exposure to environmental pollutants. The development of asthma is linked to epithelial cell damage and disruption of barrier integrity, with epithelial cell-derived cytokines driving the immune response.

Summary: This study reveals that the changes of innate lymphoid cell subsets are related to elevated IL-17A in patients with allergic rhinitis during the minimal persistent inflammation (MPI) stage. Biminkang can effectively treat MPI by regulating airway innate lymphoid cell homeostasis.

IL-10 production by ILC2s requires Blimp-1 and cMaf, modulates cellular metabolism, and ameliorates airway hyperreactivity

Emily Howard et al.

Summary: The study identified a unique subset of ILC2s, known as ILC2(10)s, which actively produce IL-10 to suppress proinflammatory ILC2 functions through autocrine and paracrine signaling, resulting in reduced lung inflammation and airway hyperresponsiveness in vivo. Furthermore, the research highlights the metabolic dependency of ILC2(10)s on the glycolytic pathway for IL-10 production, contrasting with the conventional fatty acid oxidation pathway used for proinflammatory effector functions.

Induction of IL-10-producing type 2 innate lymphoid cells by allergen immunotherapy is associated with clinical response

Korneliusz Golebski et al.

Summary: IL-10(+) ILC2s play a crucial role in modulating grass-pollen allergy, attenuating Th responses, and maintaining epithelial cell integrity. Patients with grass-pollen allergy have lower levels of IL-10(+) KLRG1(+) ILC2s, but this ability to produce IL-10 is restored after immunotherapy treatment. The mechanisms underlying this involve modifications in retinol metabolic pathway, cytokine-cytokine receptor interaction, and JAK-STAT signaling pathways in the ILCs.

Summary: Lung innate lymphoid cells (ILCs) show differences in numbers, cell surface antigen expression, and cytokine levels between neonatal and adult mice. Female mice have more lung ILCs with different antigen expression compared to males. Additionally, mouse strain impacts ILC numbers and expression in the lung.

RORα is a critical checkpoint for T cell and ILC2 commitment in the embryonic thymus

Ana C. F. Ferreira et al.

Summary: ILC2 cells can develop in the embryonic thymus from shared T cell precursors before the emergence of double-positive T cells, and migrate to mucosal tissues. The expression of ROR alpha in the thymus promotes ILC2 development while repressing T cell development. Notch signaling, BCL11B, and ROR alpha play critical roles in the co-development of T cells and ILC2 cells in the thymus.

TGFβ promotes low IL10-producing ILC2 with profibrotic ability involved in skin fibrosis in systemic sclerosis

Paoline Laurent et al.

Summary: Our study demonstrated that in SSc patients, ILC2 numbers were increased in the skin and correlated with skin fibrosis severity. The dominance of natural ILC2 over inflammatory ILC2 in SSc skin was favored by the activity of TGF-beta. TGF-beta-stimulated ILC2 showed enhanced profibrotic activity by decreasing IL10 production and increasing myofibroblast differentiation, while therapeutic intervention in a mouse model combining pirfenidone with IL10 administration effectively reduced skin infiltrating ILC2 and alleviated skin fibrosis.
