PDMS-based porous membrane for medical applications: design, development, and fabrication

The fabrication of porous membranes, which serve as cell culture substrates, is a challenging and complex process in OoC chip design. This study presents a new approach for designing and fabricating efficient porous membranes with reduced time and cost.

This review presents an overview of bioprinting techniques, their advantages and limitations, recent advancements, applications, and future prospects of the 3D-bioprinted OOC platforms, as well as current challenges and future directions.

PDMS-PEG Block Copolymer and Pretreatment for Arresting Drug Absorption in Microphysiological Devices

Summary: An effective and simple method was developed to prevent drug absorption by PDMS by incorporating a PDMS-PEG block copolymer and pretreating the drug. The method addressed the hydrophobicity and permeability of PDMS and resulted in a mean reduction of drug absorption by 91.6% under optimal conditions. The method was also demonstrated to be applicable in a PDMS-based cardiac microphysiological system, enabling more accurate drug studies.

Mechanical Properties of Soft Biological Membranes for Organ-on-a-Chip Assessed by Bulge Test and AFM

Summary: Advanced in vitro models like organ-on-a-chip can mimic specific cellular environments found in tissues, with ECM playing a key role in tissue homeostasis. Collagen-elastin (CE) membranes with mechanical properties similar to those in vivo can be produced by adjusting the CE ratio, production mode, and temperature, with stiffness ranging from several hundred kPa down to 1 kPa. Assessment using AFM shows that cell spread is lower on softer CE membranes compared to stiffer ones.

Cell-laden microgels (CLMs) are a promising tool for 3D cell culturing and on-chip generation of micro-organs, enabling the formation of tunable 3D cellular microenvironments and long-term in vitro cultures through engineering of hydrogel matrix with balanced biochemical and biophysical cues. This review provides an overview of the recent applications of CLMs in microfluidic devices for organoids formation, highlighting their roles in OOC development and future research insights.
