Vaccines for Human Schistosomiasis: Recent Progress, New Developments and Future Prospects

Adebayo J. Molehin et al.

Summary: Schistosomiasis, a neglected tropical parasitic disease caused by human blood flukes, lacks an available vaccine despite being ranked as one of the top 10 urgently needed vaccines by Science. This review highlights the current progress of schistosomiasis vaccines under clinical development and emphasizes the need for revolutionary and effective vaccine pipeline utilizing cutting-edge technologies.

Excretion patterns of Schistosoma mansoni antigens CCA and CAA by adult male and female worms, using a mouse model and ex vivo parasite cultures

Miriam Casacuberta-Partal et al.

Summary: The study found that male parasites excrete more circulating antigens than females in both the mouse model and ex vivo, and in mixed infections, serum CAA levels correlate with the number of recovered worms. In mice infected with female worms only, no viable eggs and no inflammation were seen in the livers. Additionally, CAA levels were higher than CCA levels ex vivo.

Limited efficacy of repeated praziquantel treatment in Schistosoma mansoni infections as revealed by highly accurate diagnostics, PCR and UCP-LF CAA (RePST trial)

Pytsje T. Hoekstra et al.

Summary: This study found that the efficacy of praziquantel (PZQ) treatment for schistosomiasis was overestimated when using less sensitive diagnostic methods. The results highlight the importance of using accurate diagnostic tools to monitor different treatment strategies.

In vivo efficiency of praziquantel treatment of single-sex Schistosoma three months old in mice

Ning Wang et al.

Summary: This article investigated the treatment effectiveness of praziquantel on single-sex Schistosoma japonicum. The results showed that there was no treatment efficiency observed on female schistosomes, and only at a high dose, a significant higher efficiency in reducing worm burdens was observed on male schistosomes. Moreover, the schistosomes surviving treatment still had normal reproduction potential. The study suggests the need for refining treatment strategies and developing alternative therapies to eliminate schistosomiasis.

A one-year unisexual Schistosoma mansoni infection causes pathologic organ alterations and persistent non-polarized T cell-mediated inflammation in mice

Martina Sombetzki et al.

Summary: Schistosoma spp. exhibit gonochorism and phenotypic sexual dimorphism, which is unique among trematodes. Only females mating with male schistosomes can produce highly immunogenic parasite eggs that determine the clinical symptoms of schistosomiasis. In a long-term unisexual infection experiment in mice, we found that both male and female S. mansoni can survive unpaired for one year, and this unisexual infection leads to pronounced inflammation of the liver characterized by a non-polarized Th1/Th2 immune response.

Human serum activates the tegument of female schistosomes and supports recovery from Praziquantel

Franziska Winkelmann et al.

Summary: Schistosomiasis is a devastating parasitic disease, with Schistosoma spp. surviving in the vasculature of human hosts for decades and evolving mechanisms to evade the host's immune response. Female schistosomes' resistance to Praziquantel seems to depend on active proteins in human serum, while different expression profiles of tegument-specific genes indicate different functions of female and male teguments in response to human serum.
