The ex vivo pharmacology of HIV-1 antiretrovirals differs between macaques and humans

Summary: The utility of non-human primates in evaluating antiretroviral drugs for HIV treatment and prevention is questionable due to pharmacological differences between humans and macaques. We developed an ex vivo model using tissue explants to identify differences in drug efficacy between the two species. Our results showed that high concentrations of tenofovir and maraviroc predicted anti-viral efficacy, with greater inhibitory potency observed in macaques compared to humans.

Trends in knowledge of HIV status and efficiency of HIV testing services in sub-Saharan Africa, 2000-20: a modelling study using survey and HIV testing programme data

Summary: This study examines the progress and gaps in reaching the UNAIDS 2020 target of 90% knowledge of HIV status, and the efficiency of HIV testing services in sub-Saharan Africa using data from surveys and national HIV testing reports. Knowledge of HIV status has increased from 5.7% in 2000 to 84% in 2020, with disparities among gender and age groups. In order to achieve the next UNAIDS target of 95% diagnostic coverage by 2025, addressing disparities in knowledge of status is crucial.

Pharmacokinetic/pharmacodynamic investigation of raltegravir with or without lamivudine in the context of HIV-1 pre-exposure prophylaxis (PrEP)

Summary: In this study, the combination of raltegravir and lamivudine showed better efficacy in preventing HIV infection compared to raltegravir alone, particularly providing higher protection in rectal and vaginal tissues. Drug concentrations varied in different body compartments, with concentrations persisting in rectal and vaginal tissues after discontinuation. These results suggest that raltegravir and lamivudine may be effective agents for PrEP.
