Ataxia telangiectasia mutated kinase inhibition promotes irradiation-induced PD-L1 expression in tumour-associated macrophages through IFN-I/JAK signalling pathway

Yuke Gao et al.

Summary: This study found that ionizing radiation can promote PD-L1 expression in macrophages, and ATM kinase inhibitor further enhances this effect. ATM inhibition leads to DNA damage, which activates the STING/IRF3 signaling pathway, upregulates type I interferon expression in macrophages, and ultimately leads to PD-L1 upregulation. ATM inhibition also increases CD8(+) T cell infiltration induced by IR and promotes anti-tumor efficacy in vivo.

Key chemokines direct migration of immune cells in solid tumors

Karan Kohli et al.

Summary: Immune cell infiltration into solid tumors and their interactions with chemokine receptors play a crucial role in tumor growth and the balance between effector and immunosuppressive cells. This review discusses the implications of dysregulated chemokine signaling in the tumor microenvironment and the potential for targeting chemokine signaling in cancer immunotherapy.

ATM inhibition enhances cancer immunotherapy by promoting mtDNA leakage and cGAS/STING activation

Mengjie Hu et al.

Summary: Inhibition of ATM can enhance immune checkpoint blockade therapy by promoting cytoplasmic leakage of mitochondrial DNA and activating the cGAS/STING pathway, delaying tumor growth, enhancing the efficacy of anti-PD-1 therapy, and increasing lymphocyte infiltration into the tumor microenvironment. Additionally, ATM mutations, especially nonsense mutations, can predict clinical benefits of ICB therapy.

Critical role of the CD44low CD62Llow CD8+ T cell subset in restorinng antitumor immunnity in aged mice

Yuka Nakajima et al.

Summary: CD8(+) T cells play a crucial role in antitumor immune responses, but this function is suppressed in aged individuals. The mechanism of this age-related immune decline is unclear. This study found that inefficient generation of a specific CD8(+) T cell subset may contribute to age-related resistance to PD-1 blockade, which can be overcome by strong TCR stimulation.

Low-dose targeted radionuclide therapy renders immunologically cold tumors responsive to immune checkpoint blockade

Ravi B. Patel et al.

Summary: The study demonstrates that delivering radiation semiselectively to tumors through targeted radionuclide therapy (TRT) can enhance response to immune checkpoint inhibitors (ICIs), leading to therapeutic efficacy and tumor-specific T cell memory in murine models. Combination of TRT and ICI activates production of proinflammatory cytokines, promotes tumor infiltration by CD8(+) T cells, and reduces metastases, suggesting a promising approach to improve response to ICIs for various tumor types.

Combination of radiation therapy, bempegaldesleukin, and checkpoint blockade eradicates advanced solid tumors and metastases in mice

Summary: The combination of local RT and BEMPEG showed a cooperative antitumor effect in multiple immunologically 'cold' tumor models, along with increased IL-2 receptor expression on peripheral T cells. Adding immune checkpoint blockade to RT+BEMPEG further strengthened the antitumor response in some models.

Temporal analysis of type 1 interferon activation in tumor cells following external beam radiotherapy or targeted radionuclide therapy

Justin C. Jagodinsky et al.

Summary: This study investigated the time course of STING-dependent IFN1 response following radiation in multiple murine tumor models. The results demonstrated the potential of TRT to stimulate IFN1 activation comparable to that observed with EBRT, which may be critical to the therapeutic integration of TRT with immunotherapies.
