Glucose starvation induces a switch in the histone acetylome for activation of gluconeogenic and fat metabolism genes

Summary: Acetyl-CoA is a key intermediate that plays a critical role in coordinating gene expression with metabolic state. High levels of acetyl-CoA activate genes involved in cell growth or tumorigenesis through histone acetylation, but the role of histone acetylation under low levels of acetyl-CoA is not well understood. In a yeast starvation model, we observed a significant shift in the global occupancy of histone acetylation following carbon starvation. Histone acetylation marks relocate from growth-promoting genes to genes involved in gluconeogenesis and fat metabolism. This reallocation is mediated by the histone deacetylase Rpd3p and the acetyltransferase Gcn5p.

Enzymatic transfer of acetate on histones from lysine reservoir sites to lysine activating sites

Summary: This study reveals the regulation of histone acetylation by nuclear acetyl-CoA pools and suggests a local transfer of acetate occurs during gene activation. The yeast ortholog of ACSS2, Acs2, is recruited to chromatin during quiescence exit and is preferentially associated with the most up-regulated genes, indicating the important role of acetyl group transfer in gene activation.

Long-chain fatty acyl coenzyme A inhibits NME1/2 and regulates cancer metastasis

Summary: This study reveals that the metabolic intermediate LCFA-CoA regulates cancer metastasis by inhibiting NME1 and NME2, which play important roles in cellular protein functions such as clathrin-mediated endocytosis and cancer cell migration. The inhibitory effect of LCFA-CoA on NME1 compromises its metastasis suppressor function, especially under high-fat-diet conditions.

Its NDPK activity can be inhibited by CoA through non-covalent and covalent interactions, providing a novel mechanism for the regulation of cell motility and metastasis.
