ALDH1L2 regulation of formate formyl-methionine, and ROS controls cancer cell migration and metastasis

Summary: Mitochondrial enzyme ALDH1L2 catabolizes 10-formyl-THF into CO2 and THF to produce NADPH, which is important for antioxidant defense. Decreased ALDH1L2 expression in breast cancer cells leads to increased ROS levels, formate and fMet production, and enhanced cancer cell migration via the formyl-peptide receptor (FPR). In tumor models, increased ALDH1L2 expression reduces formate and fMet accumulation, limits metastatic capacity, and reduced ALDH1L2 expression is observed in breast cancer metastases.
