Application of NAD+-Dependent Electrochemical Dehydrogenase Biosensors in Human Physiological Fluids: Opportunities and Challenges

A highly sensitive colorimetric approach based on tris (bipyridine) Ruthenium (II/III) mediator for the enzymatic detection of phenylalanine

Summary: In this study, an enzymatic assay based on Phenylalanine Dehydrogenase/NAD(+) and tris (bipyridine) Ruthenium (II/III) was developed for the accurate detection of Phenylalanine concentration. The assay showed high selectivity and achieved a detection limit of 0.33 mu M and a sensitivity of 36.6 a.u nM(-1). The method was successfully tested using biological specimens from patients with hyperphenylalaninemia, demonstrating its potential as a versatile tool for phenylalanine detection in diluted serums.

Photothermal-Contrast Method Based on In Situ Gold Nanostructure Formation for Phenylalanine Detection in Human Blood

Summary: An innovative method using photothermal contrast was demonstrated to enhance the analytical sensitivity of enzymatic phenylalanine detection at a solid surface. Gold nanoparticles produced in situ by enzymatic reaction exhibited excellent photothermal effect under green-light stimuli. Optical investigation was conducted to study the enzymatic reaction and nanoparticle formation process. The proposed photothermal-contrast approach showed a sensing range of 10-1000 μM and a detection limit of 3.6 μM for phenylalanine detection.

Structural Characterization of L-Galactose Dehydrogenase: An Essential Enzyme for Vitamin C Biosynthesis

Summary: In plants, the synthesis of ascorbic acid involves multiple metabolic pathways. This study presents the crystal structure of an L-galactose dehydrogenase from spinach, which is involved in the D-mannose/L-galactose pathway. The enzyme has similar kinetic parameters to its counterpart in camu camu, a vitamin C-rich fruit. The structure reveals insights into substrate specificity and the oxidation reaction leading to ascorbic acid accumulation. The findings contribute to a better understanding of the structure-function relationships in vitamin C synthesis enzymes.
