Degradable and Tunable Keratin-fibrinogen Hydrogel as Controlled Release System for Skin Tissue Regeneration

Summary: Biodegradable hydrogels have great potential as biomaterials for controlled-release systems in skin tissue regeneration. In this study, keratin-based hydrogels were synthesized and their properties were investigated for controllable protein delivery. The results showed that these hydrogels exhibited promising biological performance and could serve as suitable carriers for controlled protein delivery.

Wound-Healing Material with Antibacterial and Antioxidant Functions, Constructed Using Keratin, Hyperbranched Polymers, and MnO2

Summary: In this study, a low-cost and highly efficacious wound-healing material, keratin-hyperbranched polymer hydrogel-M (KHBP-M), was developed by mixing reduced keratin, hyperbranched polymer (HBP), and MnO2 nanoparticles. The composite gel showed antibacterial effects against both Gram-positive and Gram-negative bacteria and had excellent ROS-scavenging ability. In an animal model of infected wounds, the KHBP-M hydrogel treated with near-infrared light exhibited the fastest wound-healing rate, reaching 82.98% on day 15.

Assessment of the Efficacy of an LL-37-Encapsulated Keratin Hydrogel for the Treatment of Full-Thickness Wounds

Summary: This study aimed to develop a keratin-based hydrogel dressing and investigate the impact of LL-37 antimicrobial peptide delivery on treating full-thickness rat wounds. The results showed that the keratin hydrogel containing 25:75% LL-37 promoted fibroblast proliferation, adhesion, and migration, and effectively killed both Gram-negative and Gram-positive bacteria. In vivo experiments demonstrated that the L-KO25:KN75 hydrogel group had a high wound closure rate and increased microvessel density. Therefore, the LL-37-containing keratin hydrogel has the potential to improve wound closure and enhance angiogenesis for skin tissue regeneration in medical applications.

Keratose Hydrogel Drives Differentiation of Cardiac Vascular Smooth Muscle Progenitor Cells: Implications in Ischemic Treatment

Summary: Peripheral artery disease (PAD) is a common vascular disorder with limited clinical treatments. This study investigates the therapeutic potential of using keratose hydrogels to differentiate smooth muscle progenitor cells into functional smooth muscle cells (VSMCs) for PAD. The results show that keratose hydrogel can effectively drive the differentiation of VSMCs and improve blood flow and vascular densities in a mouse hindlimb ischemic PAD model.

In vivo results showed that these membranes promoted wound closure, increased angiogenesis and accelerated re-epithelization, with excessive collagen deposition at the wound sites.

Therefore, keratin biomaterials have found applications in wound healing, drug delivery, tissue engineering, and other medical fields.

The physical and biological characteristics, applications, and future research directions of BBGs are analyzed in this review paper.

The material was prepared using the freeze-gelation method and exhibited excellent liquid absorption, well-interconnected pores, good biocompatibility, and biodegradability. The results suggest that this material can reduce hemostasis time and prevent infection.
