Bioprinting a skin patch with dual-crosslinked gelatin (GelMA) and silk fibroin (SilMA): An approach to accelerating cutaneous wound healing

Summary: Clinical settings often struggle to treat large acute wounds and need alternative therapeutic approaches. Hydrogels made from natural or synthetic materials have the potential to promote wound healing. In this study, a 3D bioprinted hydrogel patch made of GelMA and SilMA was developed to accelerate acute wound healing. The GelSilMA hydrogel demonstrated improved biodegradation and mechanical properties, while the SilMA hydrogel created a moist healing environment with sufficient degradation capacity. In vivo experiments showed that the GelSilMA hydrogel patch enhanced wound closure by promoting epidermal tissue regeneration and collagen generation. Overall, the G-S hydrogel patch offers a simple and cost-effective solution for accelerating acute wound healing.

Porous composite hydrogels with improved MSC survival for robust epithelial sealing around implants and M2 macrophage polarization

Summary: This study demonstrates that injectable photo-cross-linkable porous GelMA/SilMA hydrogels encapsulating GMSCs can facilitate epithelial sealing around implants and promote M2-polarized macrophage infiltration. The hydrogels support the proliferation, survival, and spread of GMSCs, and upregulate HDs-related genes and proteins expression. The application of these hydrogels in a rat model of implant placement enhances protein expression at the implant-PIE interface and reduces HRP penetration.

Photocurable antimicrobial silk-based hydrogels for corneal repair

Summary: Corneal transplantation is the current gold standard treatment for severe corneal diseases and injuries. However, the shortage of donor tissue remains a challenge. Additionally, the difficult instillation and drainage of antibiotic eye drops in ocular surgery is an issue of concern. In this study, antimicrobial silk-based hydrogels were developed, which can be photoactivated in situ and fully adapt to the shape of corneal injuries. These hydrogels have the potential to facilitate corneal regeneration and could be applied in various soft tissue engineering fields.

A Silk Fibroin Methacryloyl-Modified Hydrogel Promoting Cell Adhesion for Customized 3D Cell-Laden Structures

Summary: Cell adhesion is crucial for tissue formation. Developing a bioink with tunable mechanical properties and enhanced cell adhesion is a challenge in tissue engineering. In this study, a printable hydrogel was developed by modifying GelMA/HAMA with SilMA. The hydrogel showed good photocuring behavior and was suitable for DLP 3D bioprinting. SilMA content could tailor the mechanical and biological properties of the hydrogel. SilMA/GelMA/HAMA hydrogel exhibited higher adhesion strength and enhanced cell actin expression. The 3D printed scaffold structure remained stable and supported cell survival and proliferation. The hydrogel has broad implications for customized 3D structures in tissue engineering.
