Novel Biological Insights Into the Common Heritable Liability to Substance Involvement: A Multivariate Genome-wide Association Study

Tabea Schoeler et al.

Summary: A common genetic liability was found to contribute to the co-occurrence of nicotine, alcohol, and cannabis consumption. Genetic correlations among 6 measures of substance dependence and frequency of use were analyzed, leading to the identification of 42 genetic variants related to substance involvement. These findings provide insights into the genetic architecture underlying the use of common psychoactive substances.

eXclusionarY: 10 years later, where are the sex chromosomes in GWASs?

Lei Sun et al.

Summary: A study conducted 10 years ago found that only 33% of GWAS results included the X chromosome, leading to recommendations to address this exclusion. However, a recent survey showed that in the NHGRI-EBI GWAS Catalog of 2021, only 25% of the reported results included the X chromosome and 3% included the Y chromosome, indicating that the exclusion persists and has become a more significant problem.

Comprehensive exploration of the genetic contribution of the dopaminergic and serotonergic pathways to psychiatric disorders

Judit Cabana-Dominguez et al.

Summary: Psychiatric disorders exhibit clinical and genetic overlap, with dopaminergic and serotonergic neurotransmission playing important roles. This study assessed the genetic contribution of these systems to eight psychiatric disorders using GWAS analyses, identifying several associated genes and gene sets.

Molecular genetics of cocaine use disorders in humans

Noelia Fernandez-Castillo et al.

Summary: Drug addiction, particularly cocaine addiction, is a major global health issue that impacts individuals, families, societies, and public health. Genetic and environmental factors play a role in the development of cocaine use disorders, with gene expression changes and epigenetic modifications contributing to the transition from drug use to abuse or dependence. Research on the genetic underpinnings of cocaine addiction, comorbidities with other psychiatric disorders, and pharmacogenetics of treatment may lead to personalized medicine approaches for effective interventions.

Anxiety Disorders: A Review

Kristin L. Szuhany et al.

Summary: Anxiety disorders have a high prevalence in the US and significantly impact the quality of life and functioning of individuals. Common symptoms include worry, fear, and panic attacks. Screening measures can assist in diagnosing anxiety disorders. Cognitive behavioral therapy and pharmacotherapy, such as SSRIs and SNRIs, are the first-line treatments for anxiety disorders.

Genome-wide association study in individuals of European and African ancestry and multi-trait analysis of opioid use disorder identifies 19 independent genome-wide significant risk loci

Joseph D. Deak et al.

Summary: Despite the significant impact of opioid use disorder (OUD), previous genome-wide association studies (GWAS) of OUD have identified a limited number of susceptibility loci. In this large-scale GWAS study, researchers optimized genetic informativeness by performing multi-trait analysis of GWAS (MTAG) with genetically correlated substance use disorders (SUDs) in individuals of European and African ancestry. The study identified several genetic associations with OUD, including variants in the OPRM1 gene and a variant in the FURIN gene, as well as 18 genome-wide significant loci associated with OUD-MTAG.

The neurobiology of human aggressive behavior: Neuroimaging, genetic, and neurochemical aspects

Francesca A. Cupaioli et al.

Summary: In modern societies, efforts are being made to improve the quality of life by better understanding the brain's influence on aggression. Neurobiological research shows that changes in specific brain regions are related to aggression, and genetic and environmental factors also play a significant role in aggressiveness.

Risk variants and polygenic architecture of disruptive behavior disorders in the context of attention-deficit/hyperactivity disorder

Summary: This study identified three genome-wide significant loci for ADHD comorbid with DBDs, with one locus on chromosome 11 found to be a strong risk locus across European and Chinese ancestries. The findings suggest an increased load of common risk variants in ADHD+DBDs compared to ADHD without DBDs, particularly in association with aggressive behavior.

Dopamine Circuit Mechanisms of Addiction-Like Behaviors

Carli L. Poisson et al.

Summary: Addiction is a complex disease characterized by substance use disorder (SUD), with primary symptoms including exaggerated substance use, social impairment, and risky behavior. Research on rodent models suggests that the striatal dopamine system plays a crucial role in both positive symptoms like increased intake and craving, and impairments in decision making, reduced sociality, and risk taking associated with SUDs. Understanding the heterogeneity of the dopamine system and related networks may lead to more targeted treatments for addiction.

Understanding the genetics and neurobiological pathways behind addiction (Review)

Alexandra Popescu et al.

Summary: The occurrence of addiction is influenced by various factors such as genetics, environment, intrinsic factors, extrinsic factors, and the nature of addictive substances. The dopamine-mesolimbic motivation-reward-reinforcement cycle is the most consistent physiological theory in addiction, with addictive substances sharing the common property of being dopamine-agonists but each having individual mechanisms and psychoactive potentials.

Irritability in Mood Disorders: Neurobiological Underpinnings and Implications for Pharmacological Intervention

Erica Bell et al.

Summary: Irritability is a common experience in both health and disease, particularly in the context of psychiatric illnesses where it is a transdiagnostic phenomenon. Despite its prevalence in mood disorders, irritability remains poorly understood and shows differences in neurobiological and neurochemical aspects between children, adolescents, and adults. Accurate definition and assessment of irritability are crucial for advancing understanding and treatment, as subjective experiences of irritability play a significant role in both health and illness.
