Long noncoding RNA Lnc-DIF inhibits bone formation by sequestering miR-489-3p

Chong Yin et al.

Summary: This study discovered an LncRNA called Lnc-DIF that inhibits bone formation by sequestering miR-489-3p through a repeat sequence. It was found that siRNA targeting Lnc-DIF could rescue bone formation in aging and ovariectomized osteoporosis mice. These findings provide potential therapeutic insights for RNA-based therapy for osteoporosis.

Targeted Intervention of NF2-YAP Signaling Axis in CD24-Overexpressing Cells Contributes to Encouraging Therapeutic Effects in TNBC

Lingmi Hou et al.

Summary: In this research, CD24(high) cells were found to be the majority of TNBC cells and were insensitive to traditional treatments. The NF2-YAP signaling axis modulated the expression of CD24 and ferroptosis suppressor protein 1 (FSP1), enhancing ferroptosis and macrophage phagocytosis. A precision targeted therapy system was designed to effectively target CD24(high) cells, inducing lysosomal escape and drug burst release through CO2 production. This system achieved dual antitumor effects.

ALX1-transcribed LncRNA AC132217.4 promotes osteogenesis and bone healing via IGF-AKT signaling in mesenchymal stem cells

Cui Zhang et al.

Summary: This study reveals that the lncRNA AC132217.4 plays a critical role in BMSC osteogenesis, promoting the development and function of osteoblasts. AC132217.4 interacts with IGF2 mRNA to regulate its expression and activate the AKT pathway, thereby controlling osteoblast maturation and function. Additionally, the splicing factors SC35 and HNRNPA1 are involved in the biogenesis of AC132217.4, while the transcription factor ALX1 regulates the expression of AC132217.7 to promote osteogenesis. In vivo over-expression of AC132217.4 significantly promotes bone healing in mice.

Polyvinylamine with moderate binding affinity as a highly effective vehicle for RNA delivery

Ye Tian et al.

Summary: Polymeric carriers, such as polyvinylamine (PVAm), offer potential advantages in RNA therapy by balancing high transfection efficacy with low toxicity. PVAm demonstrates high levels of transfection and shows promise for in vivo activity, with advantages over the gold standard polymer, polyethyleneimine (PEI), including simpler purification and lower cost. Mechanism analysis reveals the potential of PVAm for efficient RNA delivery.
