Comparison of two corticosteroid regimens on brain volumetrics in patients with Duchenne muscular dystrophy

Frontal lobe-dominant cerebral blood flow reduction and atrophy can be progressive in Duchenne muscular dystrophy

Summary: This study aimed to clarify the characteristics and progressive changes of brain image abnormalities in Duchenne muscular dystrophy (DMD). The study found that DMD patients frequently exhibit frontal lobe-dominant cerebral blood flow reduction and atrophy, and may be at risk of progressive cerebral atrophy and reduced cerebral blood flow. The findings suggest that MRI, CT, and/or brain single photon emission CT are useful for detecting brain abnormalities in adult DMD patients.

Disrupted structural connectome and neurocognitive functions in Duchenne muscular dystrophy: classifying and subtyping based on Dp140 dystrophin isoform

Summary: Neurocognitive disabilities in Duchenne muscular dystrophy (DMD) children are more likely to be significant when involving disruption of the Dp140 gene deletions. MRI data analysis techniques can provide insights into the underlying pathophysiology. This study found that motor disabilities are pronounced in the Dp140- subgroup, and IQ parameters are significantly low in all-DMD cases, with the Dp140- subgroup having the lowest scores. The structural network-based characterization of abnormalities in DMD, especially prominent in the Dp140- subgroup, suggests genotype-phenotype correlation of brain-behavior involvement in DMD children.

Effect of Different Corticosteroid Dosing Regimens on Clinical Outcomes in Boys With Duchenne Muscular Dystrophy A Randomized Clinical Trial

Summary: A double-blind, parallel-group randomized clinical trial was conducted to compare the efficacy and adverse effects of different corticosteroid regimens in boys with Duchenne muscular dystrophy. The study found that daily prednisone and daily deflazacort were more effective than intermittent prednisone for improving motor function, pulmonary function, and satisfaction with treatment over a 3-year period. There was no significant difference between the two daily corticosteroid regimens.

MRI studies have shown volumetric changes in the brains of CS patients, both during active disease and in remission.
