DNA-dependent protein kinase catalytic subunit (DNA-PKcs) drives chronic kidney disease progression in male mice

Summary: The study reveals that DNA-dependent protein kinase catalytic subunit (DNA-PKcs) drives the progression of chronic kidney disease via TAF7/RAPTOR/mTORC1 signaling. DNA-PKcs expression is significantly increased in kidney tissues of chronic kidney disease patients and male mice with kidney injuries. Inhibiting DNA-PKcs can prevent the development of chronic kidney disease in male mice, and it also preserves epithelial cell phenotype and inhibits fibroblast activation. TAF7 enhances mTORC1 activation by upregulating RAPTOR expression, promoting metabolic reprogramming in injured epithelial cells and myofibroblasts.
