Tumor-Infiltrating PD-1+Immune Cell Density is Associated with Response to Neoadjuvant Chemoradiotherapy in Rectal Cancer

Yusuke Kitagawa et al.

Summary: This study assessed the predictive and prognostic significance of pre-treatment CD8+, PD-1+, FOXP3+, CD68+, and CD163+ intratumoral or stromal cell densities in patients with rectal cancer treated with neoadjuvant chemoradiotherapy (CRT). The results showed that high intratumoral CD8+ and PD-1+ cell densities were independently associated with a good response to CRT and improved recurrence-free survival. These findings suggest that these cell densities could serve as predictive and prognostic biomarkers for rectal cancer.

Tumor-associated human dendritic cell subsets: Phenotype, functional orientation, and clinical relevance

Ioana Plesca et al.

Summary: Dendritic cells (DCs) are crucial in orchestrating anti-tumor immunity, capable of producing proinflammatory cytokines, initiating T-cell responses, and exhibiting direct cytotoxicity against tumor cells. The functionality of DCs depends on intrinsic properties and the tumor microenvironment, with immunogenic DCs promoting tumor elimination and tolerogenic DCs fostering tumor progression.

Integration, exploration, and analysis of high-dimensional single-cell cytometry data using Spectre

Thomas Myles Ashhurst et al.

Summary: The Spectre R package enables comprehensive integration and analysis of high-dimensional cytometry data from different batches or experiments. It streamlines the analytical stages of raw data pre-processing, batch alignment, integration, clustering, visualization, and statistical analysis, making it suitable for bioinformaticians and laboratory scientists. Spectre's modular design allows for scalable analysis of very large datasets generated by flow cytometry, mass cytometry, or spectral cytometry.

Genomic and transcriptomic determinants of response to neoadjuvant therapy in rectal cancer

Walid K. Chatila et al.

Summary: The incidence of rectal cancer is increasing in younger patients. Neoadjuvant therapy can avoid surgery, but determining response-related factors is important. APC mutations are associated with aggressive behavior, KRAS mutations with increased risk of relapse, and overexpression of IGF2 and L1CAM with decreased treatment response. In addition, RNA-sequencing can identify immune hot tumors with better response to neoadjuvant therapy. DNA and RNA sequencing can serve as biomarkers for treatment response in rectal cancer patients, informing non-operative treatment strategies.

Management and Outcome of Local Regrowths in a Watch-and-wait Prospective Cohort for Complete Responses in Rectal Cancer

Marit E. van der Sande et al.

Summary: The study evaluated the management and outcomes of rectal cancer patients with local regrowth in a watch-and-wait program. Most patients with regrowth were able to undergo surgical treatment with curative intent, and uncontrolled pelvic disease was very rare.

Comprehensive Phenotyping of Human Dendritic Cells and Monocytes

Florian Mair et al.

Summary: Professional antigen-presenting cells (APCs), including dendritic cells (DCs) and monocytes, play vital roles in inducing and guiding adaptive T-cell responses. Recent advancements in single-cell analysis have improved our understanding of APC subset heterogeneity, with novel phenotyping markers helping to accurately resolve functional diversity. This report provides a concise overview of traditional and recently described markers for characterizing DCs and monocytes in the human system.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

The Protein Secretome Is Altered in Rectal Cancer Tissue Compared to Normal Rectal Tissue, and Alterations in the Secretome Induce Enhanced Innate Immune Responses

Aisling B. Heeran et al.

Summary: Rectal cancer patients receive chemoradiotherapy before surgery, with varying response rates due to unknown reasons. Inflammation and the immune system play important roles in cancer treatment responses, and levels of inflammatory markers are correlated with obesity status. Irradiating rectal cancer tissue enhances immune cell anti-tumour response and does not negatively affect the tumour microenvironment's ability to induce an immune response.

Tumor-Infiltrating Cytotoxic T Cells and Tumor-Associated Macrophages Correlate With the Outcomes of Neoadjuvant Chemoradiotherapy for Locally Advanced Rectal Cancer

Yuqin Yang et al.

Summary: Tumor-infiltrating immune cells, such as CD8+ cytotoxic T lymphocytes and CD163+ tumor-associated macrophages, have been found to be predictive markers for the clinical outcomes of neoadjuvant chemoradiotherapy in locally advanced rectal cancer. High levels of CTLs were positively associated with pathological complete response, while TAMs were correlated with a poor response, suggesting their potential use in clinical decision-making for improved outcomes.

Summary: Background characterization of breast cancer includes markers such as ER, PR, HER2, and Ki67. Tumor-infiltrating lymphocytes are considered in certain BC subtypes. Additional biomarkers, such as IRF8, are being explored to improve precision in therapeutic response.
