Long-term low-dose acetylsalicylic use shows protective potential for the development of both vascular dementia and Alzheimer's disease in patients with coronary heart disease but not in other individuals from the general population: results from two large cohort studies

Thi Ngoc Mai Nguyen et al.

Summary: This study found a potential inverse association between long-term low-dose ASA use and all-cause dementia, Alzheimer's disease, and vascular dementia, especially in individuals with pre-existing coronary heart disease. Long-term use of low-dose ASA for 10 years or longer showed a strong protective effect, particularly against vascular dementia.

Molecular mechanism of acetylsalicylic acid in improving learning and memory impairment in APP/PS1 transgenic mice by inhibiting the abnormal cell cycle re-entry of neurons

Pei-Pei Guan et al.

Summary: Alzheimer's disease is a neurodegenerative disorder characterized by neuronal loss and apoptosis. Abnormal expression of cell cycle-related genes and proteins is associated with β-amyloid protein, and treatment with acetylsalicylic acid can partially restore the abnormal expression and reduce neuronal apoptosis. Acetylsalicylic acid mediates the uptake of A beta by astrocytes and activates A beta-degrading mechanisms through multiple signaling pathways, thereby improving cognitive decline.

Summary: Alzheimer's disease is a common neurodegenerative disease affecting over 35 million people worldwide, with unclear pathology and no successful treatment. However, research focusing on metabolic mechanisms similar to metabolic diseases and new optogenetic methods may provide new perspectives for studying the disease.

Aspirin in people with dementia, long-term benefits, and harms: a systematic review

Katrina A. S. Davis et al.

Summary: This review examines the evidence for the risk and benefit of long-term aspirin use in people with dementia aged over 65 years, finding a potential increased risk of bleeding events but limited by the lack of confirmatory studies and the restriction to people with Alzheimer's-type dementia. Further research in more representative groups of people with dementia is needed.

Mitochondrial dysfunction, mitophagy, and role of dynamin-related protein 1 in Alzheimer's disease

Summary: Alzheimer's disease is the most common type of dementia, and recent findings suggest that targeting mitochondria and associated proteins may be a promising alternative for preventing the disease. Mitochondrial changes, including altered structure and dynamics, play a crucial role in the progression of AD pathology.

Summary: The research suggests that low-dose aspirin use may reduce the incidence of dementia in older adults, but has less effect on MCI. Results from RCTs show inconsistent findings, indicating the need for further studies to clarify the impact of aspirin on cognitive function and dementia.

Aspirin using was associated with slower cognitive decline in patients with Alzheimer's disease

Jinyan Weng et al.

Summary: The use of aspirin was associated with slower cognitive decline over time in patients with Alzheimer's disease, but not in those with normal cognition or mild cognitive impairment. These findings indicate a potential relationship between aspirin use and cognitive decline that may vary depending on the clinical stages of cognitive impairment.
