Factors Associated With Relapse and Treatment of Myelin Oligodendrocyte Glycoprotein Antibody-Associated Disease in the United Kingdom

Chanjira Satukijchai et al.

Summary: This cohort study suggests that onset age and onset phenotype should be considered when assessing subsequent relapse risk among MOGAD patients. Among patients experiencing relapse, prednisolone, first-line immunosuppression, or a combination of those treatments may be associated with decreased risk of future relapse by approximately 2-fold.

Serum MOG-IgG in children meeting multiple sclerosis diagnostic criteria

Summary: Considering MOGAD as a distinct disease from MS, this study highlights the importance of considering MOG-IgG serology when applying MS diagnostic criteria in children. MOG-IgG seropositivity is associated with atypical features and predicts a non-MS disease course.

Summary: This study assessed the relationship between MOG-IgG titers and the clinical course of MOGAD in children and found that decreasing MOG-IgG titers were associated with a reduced risk of relapse.

Clinical Features and Risk of Relapse in Children and Adults with Myelin Oligodendrocyte Glycoprotein Antibody-Associated Disease

Alvaro Cobo-Calvo et al.

Summary: The study aimed to compare clinical features, disease course, and myelin oligodendrocyte glycoprotein (MOG) antibody dynamics between children and adults with MOG-Ab-associated disease. Children showed better recovery compared to adults, with adults having a higher risk of relapse. Children with monophasic disease became MOG-Ab negative earlier than relapsing children, but this pattern was not observed in adults.

High titers of myelin oligodendrocyte glycoprotein antibody are only observed close to clinical events in pediatrics

Allysa Lui et al.

Summary: In children with CNS demyelinating diseases, distinguishing MOG-IgG positive from negative individuals based on various characteristics at disease onset, such as age of onset, gender ratio, clinical localization, and lesion morphology, is important. MOG-IgG positive children tend to experience more severe events and exhibit more prominent optic nerve involvement. High MOG-IgG titers are closely associated with clinical events and may only be observed shortly before or after disease onset.

Summary: Intrathecal production of MOG-IgG may occur more frequently than that of AQP4-IgG, suggesting different properties of B-cell trafficking and antibody production between MOG-IgG-associated disease and AQP4-IgG-positive NMOSD.

No strong HLA association with MOG antibody disease in the UK population

Melissa Grant-Peters et al.

Summary: Improvements in assays for detecting MOG antibodies have led to the recognition of MOG-antibody-associated disease as a novel disorder. However, the etiology of MOGAD remains largely unknown. HLA analysis in MOGAD patients of European ancestry did not reveal any HLA class II associations, raising questions about the mechanism of anti-MOG antibody generation. A weak protective association of HLA-C*03:04 was observed, highlighting the need for continued efforts to better understand MOGAD genetics and pathophysiology.
