Human duodenal submucosal glands contain a defined stem/progenitor subpopulation with liver-specific regenerative potential

Vincenzo Cardinale et al.

Summary: Stem/progenitor cells with regenerative potential have been identified in the adult human duodenum. These cells can form organoids in vitro and differentiate into mature liver cells in vivo. The duodenal submucosal gland cells could potentially serve as a novel cell source for regenerative medicine of the liver.

A postnatal network of co-hepato/pancreatic stem/progenitors in the biliary trees of pigs and humans

Wencheng Zhang et al.

Summary: A network of co-hepato/pancreatic stem/progenitors exists in pigs and humans in various glands. These stem/progenitors play a crucial role in supporting hepatic and pancreatic regeneration after birth. The presence and characteristics of these stem/progenitors have been extensively studied, showing their genetic signatures and multipotentiality. Pigs are considered ideal hosts for translational and preclinical studies for cell therapies targeting hepatic and pancreatic dysfunctions.

Islet Regeneration and Pancreatic Duct Glands in Human and Experimental Diabetes

Diletta Overi et al.

Summary: There is contrasting evidence on the contribution of stem/progenitor cell populations to pancreatic regeneration in diabetes, however, the PDG compartment may play a role in this process. T2DM patients show signs of pancreatic islet regeneration and involvement of the PDG niche, and PDX1 administration could support increased endocrine pancreatic regeneration in experimental diabetes. These findings contribute to defining the role and participation of stem/progenitor cell compartments within the pancreas.

Dedifferentiation-associated inflammatory factors of long-term expanded human hepatocytes exacerbate their elimination by macrophages during liver engraftment

Chenhua Wang et al.

Summary: This study found that long-term cultured proliferating human hepatocytes (lc-ProliHH) triggered excessive macrophage activation through inflammatory response, leading to decreased transplantation efficiency. The study proposed two possible solutions, including blocking immune response with dexamethasone or maturing lc-ProliHH into organoids, to improve their transplantation efficacy.

Patch grafting of organoids of stem/progenitors into solid organs can correct genetic-based disease states

Wencheng Zhang et al.

Summary: Patch grafting is a novel strategy for transplantation of stem/progenitor organoids into porcine livers, and it has been successful in other solid organs in pigs and mice as well. The grafts consist of different types of cells and have the ability to rescue animals from genetic-based diseases. Patch grafting shows potential as a cell therapy for solid organs, but further translational studies are needed for clinical applications.

Patch grafting, strategies for transplantation of organoids into solid organs such as liver

Wencheng Zhang et al.

Summary: Epithelial cell therapies have been challenging due to inefficient transplantation methods, but patch grafting strategies have shown success in transplanting porcine GFP+ biliary tree stem/progenitor organoids/patches into livers of wild type hosts. These strategies offer potential for enabling cell therapies for solid organs.

Summary: This study demonstrated that UC-MSC therapy significantly improves liver function and long-term survival rate in patients with HBV-related DLC, without causing significant adverse events. While the impact on HCC event-free survival rate was not significant, the improvement in liver function indicators during the 48-week follow-up period was remarkable.

Cellular plasticity balances the metabolic and proliferation dynamics of a regenerating liver

Ullas Chembazhi et al.

Summary: The liver has a unique ability to regenerate in response to injuries, with a subset of hepatocytes reactivating early-postnatal-like gene expression program to proliferate and another population of metabolically active cells compensating for temporary deficits in liver function. The study also reveals the spatial and metabolic dynamics of hepatocyte proliferation after partial hepatectomy, with midlobular cells proliferating nearby while portal and central vein proximal hepatocytes remain metabolically active to preserve liver functions. Intercellular crosstalk and cellular reprogramming play a crucial role in balancing metabolic and proliferative requirements during liver regeneration.

ZNRF3 and RNF43 cooperate to safeguard metabolic liver zonation and hepatocyte proliferation

Tianliang Sun et al.

Summary: Recent research has shown that restricted chromatin accessibility in intestinal stem cells (ISCs) prevents the expression of metabolic enzymes, while fine-tuning of WNT/β-Catenin activity by ZNRF3 and RNF43 restricts proliferation in a subset of hepatocytes, preserving metabolic function. ZNRF3 and RNF43 cooperate to spatially and temporally constrain WNT/β-Catenin activity in the liver, maintaining a balance between metabolic function and hepatocyte proliferation.
