Voluntary physical activity in early life attenuates markers of fatty liver disease in adult male rats fed a high-fat diet

Farqad Abdulqader et al.

Summary: Early physical exercise can reduce the effects of high-fat diet-induced paediatric fatty liver disease, but the timing of introduction of exercise is critical. Early exercise can lower liver disease scores and prevalence, while late exercise can only alleviate some of the lesions. This is important for children, young adults and possibly into adulthood.

Generation and Characterization of CYP2E1-Overexpressing HepG2 Cells to Study the Role of CYP2E1 in Hepatic Hypoxia-Reoxygenation Injury

Nouf Alwadei et al.

Summary: In this study, a HepG2 cell line with stable overexpression of CYP2E1 was generated to investigate its role in hypoxia/reperfusion injury. The results showed that the high expression of CYP2E1 in the cells led to increased reactive oxygen species production and cell death in the hypoxia/reperfusion model, suggesting a detrimental role of CYP2E1 in hepatic ischemia/reperfusion injury.

CS27109, A Selective Thyroid Hormone Receptor-β Agonist Alleviates Metabolic-Associated Fatty Liver Disease in Murine Models

Shengjian Huang et al.

Summary: A novel oral and liver-targeted THR-beta agonist, CS27109, showed comparable efficacy with MGL3196 in the treatment of metabolic disorders such as dyslipidemia and steatohepatitis. CS27109 demonstrated pronounced activity and selectivity to THR-beta and favorable pharmacokinetic properties. In animal models, CS27109 effectively reduced serum cholesterol and lipid levels, alleviated liver inflammation and steatosis, and exhibited tolerable toxicity to the heart.

Diagnostic efficiency on ultrasound shear wave elastography in evaluation of steatosis severity for non-alcoholic fatty liver disease: a rat model

Yuhui Wu et al.

Summary: The influence of steatosis on liver stiffness measured by shear wave elastography (SWE) in a rat model with NAFLD was analyzed in this study. SWE showed potential in grading steatosis in the absence of fibrosis. However, its effectiveness in distinguishing different stages of steatosis was limited.

Digital pathology and artificial intelligence in translational medicine and clinical practice

Vipul Baxi et al.

Summary: Recent technological advancements have enabled the development of digital pathology and AI-based solutions for quantitative pathologic assessments, revolutionizing disease diagnosis and drug development. These innovations provide valuable opportunities in immuno-oncology for deciphering complex pathophysiology and discovering novel biomarkers, while also supporting practitioners in selecting the most appropriate treatment based on patient profiles. The integration of AI-powered analysis tools enhances the traditional role of pathologists in delivering accurate diagnoses and assessing biomarkers, with potential applications in translational medicine and clinical settings.

The Role of Neuregulin-1 in Steatotic and Non-Steatotic Liver Transplantation from Brain-Dead Donors

Marc Mico-Carnero et al.

Summary: This study investigated the role of Neuregulin-1 (NRG1) in liver transplantation from brain death donors (DBD). The results showed that NRG1 and p21-activated kinase 1 (PAK1) levels increased in both steatotic and non-steatotic grafts from DBDs. The protective effects of NRG1 and PAK1 were associated with increased levels of VEGFA and IGF1, respectively. However, exogenous NRG1 was not an appropriate strategy for liver grafts from DBD.

Senescent hepatic stellate cells promote liver regeneration through IL-6 and ligands of CXCR2

Naiyuan Cheng et al.

Summary: Senescent hepatic stellate cells play a critical role in liver regeneration by secreting IL-6 and CXCR2 ligands, which activate multiple signaling pathways to stimulate hepatocyte proliferation. The matricellular protein CCN1 is responsible for inducing senescence in hepatic stellate cells and promoting liver regeneration.

Circulating CXCL10 and IL-6 in solid organ donors after brain death predict graft outcomes

Lorenzo Piemonti et al.

Summary: The study found that circulating CXCL10 and IL-6 levels in donor after brain death can predict graft outcomes. Several factors affect the concentrations of IL-6 and CXCL10 in donors, and these cytokines act as independent predictors of Immediate Graft Function. Serum IL-6 and CXCL10 in donors can provide additional incremental prediction of graft outcomes for recipients.

Contemporary strategies to assess and manage liver donor steatosis: a review

Christine Tien et al.

Summary: Severe liver allograft (>60%) steatosis is associated with inferior outcomes, while mild (<30%) steatosis is not. There is ongoing debate regarding the safe utilization of grafts with moderate (30-60%) steatosis. Liver biopsy is the gold standard for evaluating steatosis, but noninvasive methods are showing promise. Machine perfusion may be a way to optimize steatotic grafts before transplantation.

New Insights Into the Role of Autophagy in Liver Surgery in the Setting of Metabolic Syndrome and Related Diseases

Ana Isabel Alvarez-Mercado et al.

Summary: Visceral obesity is closely related to metabolic syndrome and diabetes, contributing to a higher risk of post-surgical complications in liver surgery. Metabolic diseases like NAFLD and diabetes exacerbate hepatic I/R injury, leading to poorer outcomes after liver surgery. Autophagy plays a crucial role in these conditions and may serve as an effective therapeutic target to improve postoperative results for patients with metabolic diseases undergoing liver surgery.
