Epigenetic basis of oncogenic-Kras-mediated epithelial-cellular proliferation and plasticity

Preetish Kadur Lakshminarasimha Murthy et al.

Summary: Oncogenic Kras induces cell proliferation and tumor development in epithelial tissues. The AP-1 factor, guided by the mSWI/SNF complex, controls gene expression necessary for tumor transformation. Cell-type-specific transcription factors can also retain the epigenetic memory of the tumor's cell of origin.

The Fra-1/AP-1 Oncoprotein: From the Undruggable Transcription Factor to Therapeutic Targeting

Laura Casalino et al.

Summary: Cancer metastasis largely relies on the phenotypic changes and genetic reprogramming orchestrated by transcription factors. Fra-1 is a crucial component in the metastatic process and represents an ideal therapeutic target. However, the structural features of Fra-1 pose challenges in designing inhibitory drugs for this nuclear oncoprotein.

SP600125 Enhances Temperature-Controlled Repeated Thermal Stimulation-Induced Neurite Outgrowth in PC12-P1F1 Cells

You-Ran Luo et al.

Summary: This study evaluated the mechanism of temperature-controlled repeated thermal stimulation (TRTS)-mediated neuronal differentiation. The researchers found that SP600125, a JNK inhibitor, can enhance TRTS-induced neuritogenesis by attenuating the negative feedback loop of BMP signaling. This study is important for the future development of regenerative neuromedicine.

JUNB-FBXO21-ERK axis promotes cartilage degeneration in osteoarthritis by inhibiting autophagy

Zhiming Lin et al.

Summary: The study revealed that suppressing FBXO21 can alleviate cartilage degeneration in OA and promote chondrocyte growth and synthesis while reducing cell death. Furthermore, FBXO21 inhibits autophagy by interacting with ERK, thus accelerating the progression of cartilage degeneration in OA.

Blocking Fra-1 sensitizes triple-negative breast cancer to PARP inhibitor

Dandan Song et al.

Summary: The overexpression of AP-1 member Fra-1 in TNBC has a significant impact on tumor progression and resistance to treatment. The interaction between Fra-1 and PARP1 affects the efficacy of olaparib treatment, with PARP1 downregulating Fra-1 and reducing AP-1 transcriptional activity. Inhibiting Fra-1/AP-1 can sensitize non-BRCA-mutated TNBC cells to olaparib treatment.

FOSL1 promotes metastasis of head and neck squamous cell carcinoma through super-enhancer-driven transcription program

Ming Zhang et al.

Summary: This study revealed that FOSL1 is significantly upregulated in HNSCC tumor tissues and promotes tumorigenicity and metastasis by establishing super-enhancers at cancer stemness and pro-metastatic genes. The abundance of FOSL1 is positively associated with SNAI2, and high expression levels of both genes are correlated with shorter disease-free survival. Inhibiting FOSL1 with the inhibitor SR11302 significantly suppressed tumor growth and lymph node metastasis in a patient-derived xenograft model, suggesting FOSL1 as a potential therapeutic target for HNSCC metastasis.

JUNB suppresses distant metastasis by influencing the initial metastatic stage

Juliane Wutschka et al.

Summary: The loss of JUNB in the tumor microenvironment has significant impacts on breast cancer progression and metastasis, leading to increased infiltration of cells and immune cells which promote metastasis. Findings suggest that JUNB plays a crucial role in suppressing distant metastasis in cancer.

CDK4/6 inhibition reprograms the breast cancer enhancer landscape by stimulating AP-1 transcriptional activity

April C. Watt et al.

Summary: Goel and colleagues demonstrate that CDK4/6 inhibitors induce global chromatin changes in breast cancer cells by activating a wide range of enhancers, leading to biological effects such as cell differentiation and apoptotic evasion. This study provides insights into the molecular mechanisms underlying the effects of CDK4/6 inhibition and highlights the potential for further development of CDK4/6 inhibitors.
