Spinal pain processing in arthritis: Neuron and glia (inter)actions

Hans-Georg Schaible et al.

Summary: Diseases of joints lead to chronic pain due to the persistent hyperexcitability of the peripheral and central nociceptive system. This article focuses on the mechanisms of spinal sensitization caused by arthritis, involving glutamate receptors, neuropeptides, prostaglandins, and proinflammatory cytokines. Activation of spinal glia cells is coincident with mechanical hyperalgesia, which can be attenuated by intrathecal interventions. The interaction between neurons and glia cells, as well as the suppression of spinal sensitization and inflammatory process, contribute to the phenotype of arthritis pain.

Rheumatoid arthritis: advances in treatment strategies

Peeyush Prasad et al.

Summary: Rheumatoid arthritis is an autoimmune disease that causes severe joint and bone damage. Treatment strategies include medication and personalized therapy. Conventional medication along with glucocorticoid is suitable for early cases, while a combination of conventional and targeted medication is effective for seronegative patients with moderate to high activity. Adverse events call for exploration of new treatment methods.

Nanomedical approaches in the realm of rheumatoid arthritis

Andrei-Flavius Radu et al.

Summary: Rheumatoid arthritis (RA) is an autoimmune inflammatory disorder characterized by bone and cartilage damage in the synovium, resulting in joint impairment and increased mortality rate. Despite recent improvements in RA treatment, only a small fraction of patients achieve clinical remission without ongoing immunosuppressive drugs. Research on RA nano therapies has increased due to their potential advantages over traditional systemic treatments. Nanotherapeutic techniques, specifically drug delivery nano systems, offer new approaches to overcome the limitations of existing treatments by precisely delivering therapeutic drugs to affected sites.

The role of dendritic cells and their immunometabolism in rheumatoid arthritis

Yuichi Suwa et al.

Summary: Dendritic cells (DCs) have important roles in the pathogenesis of rheumatoid arthritis (RA), characterized by chronic synovitis and joint destruction. Conventional dendritic cells (cDCs) in the synovium show enhanced migratory capacities and T cell activation compared to peripheral blood cDCs, while plasmacytoid dendritic cells in RA may have tolerogenic effects. Monocyte-derived dendritic cells (moDCs) in the synovium induce expansion of T-helper 17 cells and proinflammatory cytokine production. Recent studies suggest a link between proinflammatory hypoxic environments in the synovium and metabolic reprogramming of DCs. Targeting the immunometabolism of DCs could be a potential therapeutic approach in RA.

Neutrophils Lose the Capacity to Suppress T Cell Proliferation Upon Migration Towards Inflamed Joints in Juvenile Idiopathic Arthritis

Sabine Arve-Butler et al.

Summary: This study found that neutrophils in the synovial fluid of children with oligoarticular juvenile idiopathic arthritis (JIA) are unable to effectively suppress the proliferation of activated T cells. This may be due to decreased neutrophil reactive oxygen species (ROS) production and alterations in proteins related to cell-cell contact and inflammation. The lack of T cell suppression by neutrophils in the synovial fluid may contribute to local inflammation and autoimmune reactions in the JIA joint.

Novel Insights into the Molecular Mechanisms Involved in the Neuroprotective Effects of C-Phycocyanin against Brain Ischemia in Rats

Javier Marin-Prida et al.

Summary: This study reveals the neuroprotective effects of C-Phycocyanin (C-PC) against brain ischemia by inhibiting NADPH oxidase expression and improving neuroinflammation. C-PC shows a dose-dependent reduction in infarct volume and neurological deficit in rats with cerebral ischemia/reperfusion (I/R), and also improves exploratory activity. In addition, C-PC downregulates the expression of pro-inflammatory genes and upregulates immune suppressive genes in hypoperfused brain areas. It also decreases chronic neuronal death in the hippocampus of hypoperfused rats.
