Single-Cell RNA Sequencing Identifies Intra-Graft Population Heterogeneity in Acute Heart Allograft Rejection in Mouse

Yunhua Tang et al.

Summary: In this study, single-cell RNA sequencing technology was used to analyze graft-infiltrating cells in a mouse heart transplant model. The results identified different cell types and states associated with acute rejection, including specific macrophage and endothelial cell populations. The upregulation of Ubiquitin D in rejection was also validated as a potential biomarker for rejection in the clinic.

Donor Macrophages Modulate Rejection After Heart Transplantation

Benjamin J. Kopecky et al.

Summary: This study investigates the role of donor immune cells in heart transplantation and identifies donor CCR2(+) macrophages as key mediators of allograft rejection. Deletion of MYD88 signaling in donor macrophages suppresses rejection and extends allograft survival. These findings highlight the potential for interventions targeting donor immune cells in heart transplantation.

Single-Cell RNA sequencing reveals immune cell dynamics and local intercellular communication in acute murine cardiac allograft rejection

Zhang Chen et al.

Summary: This study investigated the immune cell dynamics and local intercellular communication of acute cardiac allograft rejection using single-cell RNA sequencing. The results revealed the functional characteristics and differentiation trajectory of various alloreactive cell subpopulations, and suggested that the CXCR3 pathway may serve as a potential therapeutic target for transplant rejection.

Single-cell transcriptomic identified HIF1A as a target for attenuating acute rejection after heart transplantation

Yuan Chang et al.

Summary: Acute rejection (AR) is a significant contributor to graft failure post-heart transplantation, and the regulation of immune metabolism, particularly the increased glucose metabolism of cardiac macrophages in AR patients, has become a new focus for immunosuppressive drug development. Through single-cell RNA sequencing, it was found that during AR, cardiac resident macrophages were replaced by monocytes and proinflammatory macrophages, which exhibited higher antigen presentation and inflammatory recruitment capabilities, as well as glycolysis. The increase in glycolysis in monocytes/macrophages was primarily regulated by HIF1A, and inhibition of HIF1A could reduce inflammatory cells infiltration in AR.

Untangling Local Pro-Inflammatory, Reparative, and Regulatory Damage-Associated Molecular-Patterns (DAMPs) Pathways to Improve Transplant Outcomes

Gaelen K. Dwyer et al.

Summary: Inflammatory responses following solid organ transplantation are vital for the immune system to react to foreign antigens, but excessive or insufficient DAMPs can lead to negative outcomes. While DAMPs play a role in initiating anti-inflammatory and reparative functions, both their abundance or deficiency can impact tissue repair or the establishment of homeostasis.

Innate allorecognition in transplantation

Khodor I. Abou-Daya et al.

Summary: Successful allogeneic transplantation relies on suppressing activation of the adaptive immune system. While current immunosuppressive therapy targets T and B cells to prevent rejection, recent research has highlighted the crucial role of the innate immune system in initiating the allo-immune response. Understanding innate allorecognition better could lead to improved treatment strategies and prolong allograft survival.

Summary: UCell is an R package designed for evaluating gene signatures in single-cell datasets. It offers robust signature scores based on the Mann-Whitney U statistic, requiring less computing time and memory compared to other methods. UCell can handle large datasets in minutes and can interact directly with Seurat objects.
