An integrated cell atlas of the lung in health and disease

Lisa Sikkema et al.

Summary: Single-cell technologies are used to study human tissues, but individual studies have limitations in terms of sample size and cell type definitions. The integrated Human Lung Cell Atlas (HLCA) combines 49 datasets from 486 individuals to create a comprehensive atlas of the human respiratory system. The HLCA provides new annotations for different cell types and allows for the identification of gene modules associated with demographic variables and changes in gene expression.

Challenging the Ex Vivo Lung Perfusion Procedure With Continuous Dialysis in a Pig Model

Julien De Wolf et al.

Summary: This study investigated the effects of normothermic ex vivo lung perfusion (EVLP) on the physiological, metabolic, and genome-wide response in a pig model. The results showed that the management of EVLP perfusate by periodic replacement and continuous dialysis had no significant effect on lung function nor on the gene expression profiles ex vivo, suggesting that the accumulation of dialyzable cell products does not significantly alter the lung cell response during EVLP, which has implications for EVLP management in the clinic.

Prediction of donor related lung injury in clinical lung transplantation using a validated ex vivo lung perfusion inflammation score

Andrew T. Sage et al.

Summary: This study developed a protein-based assay using IL-6 and IL-8 levels in perfusate to assess lung injury during EVLP, accurately predicting transplant suitability and recipient outcomes. The inflammation score in conjunction with Delta PO2 successfully identified EVLP transplants likely to have favorable recipient outcomes, improving organ utilization rates and patient outcomes.

Ex vivo lung perfusion

Tiago N. Machuca et al.

Summary: Ex vivo lung perfusion has emerged as a safe method for preserving and reassessing marginal lung grafts, expanding the donor pool and increasing global lung transplant activity. Additionally, it serves as a platform for advanced diagnostics, enabling specific targeted or personalized treatments.

The potential of ex vivo lung perfusion on improving organ quality and ameliorating ischemia reperfusion injury

Jasper Iske et al.

Summary: Allogeneic lung transplantation is the treatment of choice for advanced lung diseases, but ischemia reperfusion injury remains a crucial issue during the transplant process. Ex vivo lung perfusion technique can reduce the incidence of primary graft dysfunction and improve organ quality.

Transcriptomic investigation reveals donor-specific gene signatures in human lung transplants

Cristina Baciu et al.

Summary: Gene expression profiling revealed distinguishable biological transcriptome signatures in DBD and DCD lungs, with DBD lungs showing upregulation of inflammatory pathways and DCD lungs displaying transcriptome signatures associated with cell death. Specific drug targets for rehabilitating DBD lungs were identified, and activation of certain signaling pathways were observed in EVLP-treated DBD lungs.

Actin cytoskeleton dynamics during mucosal inflammation: a view from broken epithelial barriers

Susana Lechuga et al.

Summary: Disruption of epithelial barriers is a key pathogenic event of mucosal inflammation, attributed to abnormal structure and permeability of epithelial junctions. The altered biochemical and physical environment of inflamed tissues affects the dynamics of the actomyosin cytoskeleton, leading to barrier disruption.
