The interplay of autophagy and oxidative stress in the pathogenesis and therapy of retinal degenerative diseases

Kun-Che Chang et al.

Summary: Oxidative stress caused by intracellular reactive oxygen species (ROS) production is closely associated with the pathogenesis of ocular diseases. Autophagy, a self-clearance pathway, plays an antioxidant role by regulating cellular ROS levels. However, excessive autophagy may lead to cell death. This review summarizes the mechanisms of interaction between ROS and autophagy and their roles in several ocular diseases.

Cell-to-Cell Communications in Alcohol-Associated Liver Disease

Natalia A. Osna et al.

Summary: This review focuses on the communication between liver parenchymal and non-parenchymal cells induced by alcohol, which plays a significant role in the development of liver injury. Various mechanisms are involved in the information exchange between different cell types, and this exchange may promote the progression of end-stage liver disease. The review highlights the activation of non-parenchymal cells, liver macrophages, and hepatic stellate cells by extracellular vesicles derived from alcohol-exposed hepatocytes. Additionally, it discusses the crosstalk between cell death pathways and inflammasome activation in alcohol-activated cells, as well as the role of non-inflammatory factors, sinusoidal pressure, and hepatic arterialization in alcohol-induced hepatic fibrogenesis.

Sirtuin 3 deficiency exacerbates diabetic cardiomyopathy via necroptosis enhancement and NLRP3 activation

Shu Song et al.

Summary: The study showed that reduced expression of SIRT3 in diabetic cardiomyopathy worsens cardiac dysfunction, myocardial injury, ROS accumulation, necroptosis, and inflammation, ultimately exacerbating the condition. This suggests SIRT3 could be a potential molecular target for preventing and treating diabetic cardiomyopathy.

Hepatocyte pyroptosis and release of inflammasome particles induce stellate cell activation and liver fibrosis

Susanne Gaul et al.

Summary: The study found that hepatocyte pyroptosis, caused by NLRP3 inflammasome activation, leads to the release of inflammasome proteins and exacerbates fibrogenesis. Inhibition of caspase-1 and gasdermin D activation can prevent pyroptosis. The released inflammasomes are taken up by hepatic stellate cells, promoting inflammation and fibrosis. These findings suggest a novel mechanism for liver injury and potential therapeutic targets for chronic liver diseases.

Therapeutic potential of the target on NLRP3 inflammasome in multiple sclerosis

Shuai Shao et al.

Summary: Inflammasomes are multi-protein complexes that regulate the maturation and secretion of IL-1 beta and IL-18 in response to PAMPs or DAMPs in host cells. The NLRP3 inflammasome is well-understood in molecular mechanisms, serving as a promising therapeutic target in immune-related disorders such as multiple sclerosis (MS). Investigation of NLRP3 activation mechanisms in MS and experimental models may lead to new therapeutic strategies for the disease.

A long way to go: caspase inhibitors in clinical use

Shanel Dhani et al.

Summary: Caspases are a family of evolutionarily conserved cysteine-dependent proteases involved in vital cellular processes. Dysregulation of caspase-mediated apoptosis and inflammation has been linked to the pathogenesis of various diseases, leading to the development of caspase inhibitors as potential therapeutic tools. However, challenges such as inadequate efficacy, poor target specificity, and adverse side effects have limited the progress of these inhibitors in clinical trials. Further studies on understanding caspase function in disease models are essential to successfully develop therapeutic caspase inhibitors for various pathologies.
