Seroprevalence of Binding and Neutralizing Antibodies against 39 Human Adenovirus Types in Patients with Neuromuscular Disorders

Patrick Julian Klann et al.

Summary: In this study, the seroprevalence of various adenoviruses was tested in patients with neuromuscular disorders (NMDs) and healthy controls to identify suitable viral vectors for gene replacement therapy for NMDs. The results showed that patients with NMDs had similar levels of antibodies against adenoviruses compared to healthy individuals. HAdV-A31, HAdV-B16, HAdV-B34, HAdV-B35, HAdV-D8, HAdV-D37, HAdV-D48, HAdV-D73, HAdV-D74, HAdV-D75, and HAdV-D80 were identified as promising candidates for future vector development due to their low antibody prevalence.

The Adenovirus Vector Platform: Novel Insights into Rational Vector Design and Lessons Learned from the COVID-19 Vaccine

Erwan Sallard et al.

Summary: The adenovirus vector platform is an efficient toolbox for gene therapy, virotherapy, and vaccine development. The adenovirus genome and capsids can be modified using highly efficient techniques, and high-titer vectors can be produced to meet current needs. In recent years, the adenovirus vector platform has been extensively studied for the development of COVID-19 vaccines. The worldwide deployment of these vaccines has increased our knowledge on virus-host interactions and the need for further improvement.

Cell entry and innate sensing shape adaptive immune responses to adenovirus-based vaccines

Sonia Marquez-Martinez et al.

Summary: Nonreplicating adenovirus-based vectors are effective prophylactic vaccines against infectious viral diseases, inducing protective cellular and humoral responses. Differences in cellular entry or endosomal escape mechanisms contribute to differences in innate immune sensing between adenovirus species. Understanding these early events is critical for developing vaccines that elicit effective and durable adaptive immune responses with acceptable reactogenicity.

COVID-19 Vaccines: Adenoviral Vectors

Catherine Jacob-Dolan et al.

Summary: This review evaluates the use of adenovirus vector platforms in response to the COVID-19 pandemic. Ad vectors have been used in the past for vaccines against other viruses, but never on such a large scale. Several different serotypes of Ad encoding SARS-CoV-2 Spike have been tested and found to be effective against COVID-19. As vaccine rollouts continue, we will continue to learn about this vaccine platform.

A systematic review of vaccine-induced thrombotic thrombocytopenia in individuals who received COVID-19 adenoviral-vector-based vaccines

Mostafa H. Elberry et al.

Summary: This systematic review investigates the demographics, commonalities, and prognosis of thrombotic response after receiving COVID-19 Adenoviral-Vector Based Vaccines. The study includes cases of thrombotic events following vaccination with ChAdOx1 nCoV-19 and Ad26.COV2.S vaccines. The findings suggest that young individuals, particularly females, may be more susceptible to vaccine-induced thrombosis. The most common presentations of thrombotic events include deep vein thrombosis, cerebral venous sinus thrombosis, and pulmonary embolism.

ICTV Virus Taxonomy Profile: Adenoviridae 2022

Maria Benko et al.

Summary: The family Adenoviridae consists of non-enveloped viruses with linear dsDNA genomes and medium-sized icosahedral capsids. Adenoviruses have been found in vertebrates from fish to humans. The family is divided into six genera, each of which is more prevalent in certain animal groups. The outcome of infection can range from subclinical to lethal disease.

Understanding thrombosis with thrombocytopenia syndrome after COVID-19 vaccination

Alessandra Buoninfante et al.

Summary: This article summarizes the discussion of an EMA workshop on the epidemiology, clinical presentation, and biology of thrombosis with thrombocytopenia syndrome after adenovirus vector COVID-19 vaccination. International regulators, scientists, and developers have reached a general agreement on the steps needed to improve the understanding of this new syndrome, including enhancing post-vaccination surveillance activities in low and middle income countries and investigating potential genetic predisposition factors.

Summary: The study found that severe pneumonia incidence was higher in HAdV-7-infected individuals compared to those infected with HAdV-3; HAdV-7 replicated more efficiently ex vivo than HAdV-3; Children infected with HAdV-7 showed higher inflammatory responses, and the detection of adenovirus DNA in serum samples can help assess the risk of severe disease.

Insights in ChAdOx1 nCoV-19 vaccine-induced immune thrombotic thrombocytopenia

Andreas Greinacher et al.

Summary: The mechanism underlying vaccine-induced immune thrombotic thrombocytopenia (VITT) primarily involves the formation of PF4/vaccine complexes and the proinflammatory environment stimulated by the vaccine, leading to the production of high-titer anti-PF4 antibodies in the body and consequent thrombosis.

Thrombotic Thrombocytopenia after ChAdOx1 nCov-19 Vaccination

Andreas Greinacher et al.

Summary: After vaccination with ChAdOx1 nCov-19, rare immune thrombotic thrombocytopenia can occur due to platelet-activating antibodies against PF4, resulting in various thrombotic events. Most patients in this case series were women, with fatal outcomes such as cerebral venous thrombosis and intracranial hemorrhage. Additional studies are needed to further investigate and understand this phenomenon.

Interactions of adenoviruses with platelets immune thrombotic thrombocytopenia syndrome

Paolo Gresele et al.

Summary: The COVID-19 pandemic has had a significant impact on global health and economy, with vaccination being the primary method of controlling infection. Unexpected thrombotic events have occurred in individuals who received the AstraZeneca or Johnson&Johnson adenovirus vector-based vaccines during the vaccination campaign. Studies suggest that an acute immunological reaction, potentially triggered by platelet activation, may be responsible for the novel syndrome of vaccine-associated thrombotic thrombocytopenia.
