Age-associated B cells are heterogeneous and dynamic drivers of autoimmunity in mice

Kevin M. Nickerson et al.

Summary: Age-associated B cells (ABCs) are a type of memory B cell expressing the transcription factor T-bet, formed under inflammatory conditions. In systemic lupus erythematosus (SLE), ABC frequency is correlated with disease and they are considered the source of autoantibody-secreting cells. The heterogeneity of ABCs and their relationship to other B cell subsets such as plasmablasts is unclear. The study shows that ABCs are dynamic and can be plasmablast precursors or undergo cycles of reactivation. Deletion of CD11c-expressing B cells reveals a direct role for ABC-like B cells in lupus pathogenesis.

Single-cell RNA sequencing analyses: interference by the genes that encode the B-cell and T-cell receptors

Timothy Sundell et al.

Summary: In the study, it was found that removing the gene sequences that encode B- and T-cell receptors before unsupervised clustering of single-cell RNA sequencing data resulted in clusters that represented biologically meaningful subsets. Conversely, if these genes were not removed, the resulting clusters did not represent biologically meaningful subsets.

Clonally expanded B cells in multiple sclerosis bind EBV EBNA1 and GlialCAM

Tobias Lanz et al.

Summary: Multiple sclerosis (MS) is an autoimmune disease where autoreactive lymphocytes attack the central nervous system. Epstein-Barr virus (EBV) infection has been linked to MS, with molecular mimicry between EBV nuclear antigen 1 (EBNA1) and central nervous system protein GlialCAM. EBNA1 immunization exacerbates disease in MS mouse models and anti-EBNA1 and anti-GlialCAM antibodies are prevalent in MS patients, providing a mechanistic link between MS and EBV and guiding the development of new MS therapies.

Single-cell RNA-seq reveals cell type-specific molecular and genetic associations to lupus

Richard K. Perez et al.

Summary: By using mux-seq technology, we conducted extensive single-cell RNA sequencing on peripheral blood cells from SLE patients and healthy individuals. We found increased expression of type 1 interferon-stimulated genes in monocytes, reduced naive CD4(+) T cells, and expanded cytotoxic CD8(+) T cells in SLE patients. The cell type-specific expression features could predict the case-control status and classify patients into two molecular subtypes. Furthermore, we linked SLE-associated genetic variants to cell type-specific expression using genotyping data.

Integrated single cell and spatial transcriptomics reveal autoreactive differentiated B cells in joints of early rheumatoid arthritis

Uta Hardt et al.

Summary: This study investigated the presence of differentiated B cells in the joint tissue of patients with Rheumatoid Arthritis (RA) at the onset of the disease. The findings showed that different types of B cells, including memory and plasma cells, were present regardless of the patients' autoantibody status. The study also revealed the mechanisms of local B cell maturation and plasma cell survival in the tissue. Furthermore, the research demonstrated the presence of local autoreactive plasma cell differentiation in joint biopsies captured from untreated early RA.

An introduction to spatial transcriptomics for biomedical research

Cameron G. Williams et al.

Summary: Single-cell transcriptomics (scRNA-seq) has become essential in biomedical research, especially in the fields of developmental biology, cancer, immunology, and neuroscience. However, most available protocols require intact and viable cells, limiting the scope of study and destroying spatial context. This review introduces different methods of spatial transcriptomics and discusses their features, such as tissue area, spatial resolution, and gene profiling capabilities. It also provides guidance on platform selection, data analysis, and integration with existing scRNA-seq data, aiming to help researchers utilize spatial transcriptomics in their own biomedical research.

Single-cell approaches to investigate B cells and antibodies in autoimmune neurological disorders

Alicia Zou et al.

Summary: Autoimmune neurological disorders are characterized by the presence of autoantibodies against neurological antigens, but the characteristics of a single antibody cannot be definitively assigned due to the heterogeneity of patient samples. Single-cell technologies have advanced the understanding of these disorders by revealing abnormalities in tolerance mechanisms and providing insight into B cells responsible for autoantibody production.
