A cell-free antigen processing system informs HIV-1 epitope selection and vaccine design

Summary: Distinct CD4(+) T cell epitopes associated with spontaneous control of HIV-1 replication were analyzed using a cell-free antigen processing system. Factors influencing epitope selection were identified, including protein stability and solvent accessibility. Efficient processing of some HIV-1 antigens was associated with limited CD4(+) T cell responses, while inefficient processing was observed for certain protective epitopes. This in vitro processing system provides insights into epitope selection for HIV-1 and non-HIV-1 antigens.
