Toll-like Receptor 9 Induced Dendritic Cell Activation Promotes Anti-Myeloperoxidase Autoimmunity and Glomerulonephritis

Sharon L. Ford et al.

Summary: This study investigates the role of Toll-like receptor 9 (TLR9) in murine experimental anti-MPO glomerulonephritis. TLR9 stimulation enhances dendritic cell activation, promotes systemic anti-MPO autoimmunity and T cell recall responses, and exacerbates kidney injury.

Shanze Chen et al.

Summary: Macrophages are vital to the innate immune system and play a central role in various immune tasks. They possess polarization settings that are crucial for immune regulation, and their clinical importance in cancer and other diseases is increasingly recognized. Macrophages have a significant impact on processes such as angiogenesis, tumor cell proliferation, metastasis, and immune suppression.

Increased Circulating Cell-Free DNA in Eosinophilic Granulomatosis With Polyangiitis: Implications for Eosinophil Extracellular Traps and Immunothrombosis

Teppei Hashimoto et al.

Summary: In antineutrophil cytoplasmic antibody-associated vasculitis (AAV), levels of cfDNA are increased and associated with disease activity. Among AAV patients, those with eosinophilic granulomatosis with polyangiitis (EGPA) had the highest levels of cfDNA, and in vitro experiments showed that EETs are more stable against DNase and provide a scaffold for platelet adhesion.

Identification of Hub Biomarkers and Immune-Related Pathways Participating in the Progression of Antineutrophil Cytoplasmic Antibody-Associated Glomerulonephritis

Meng-Di Xia et al.

Summary: This study identifies key biomarkers and immune-related pathways in the progression of ANCA-associated glomerulonephritis (ANCA-GN). B cells and monocytes are found to be closely associated with the pathogenesis of ANCA-GN, and five hub genes (CYP3A5, SLC12A3, BGN, TAPBP, and TMEM184B) may play a role in the progression of ANCA-GN through immune-related signal pathways.

Summary: Neutrophil extracellular traps (NETs) are a new form of cell death, not limited to neutrophils, but also found in other innate cells. This mechanism enhances host defense by trapping bacteria with DNA webs, but it can also cause tissue damage and is associated with various diseases.

CD163 and CD206 expression define distinct macrophage subsets involved in active ANCA-associated glomerulonephritis

Joop P. Aendekerk et al.

Summary: In this multicenter retrospective cohort study, the clinicopathological significance of CD206 and CD163 in anti-neutrophil cytoplasmic antibody (ANCA) mediated-vasculitis (AAV) with glomerulonephritis (ANCA GN) was investigated. The study confirmed that CD206+ and CD163+ macrophages play prominent roles in the immunopathology of ANCA GN. Different macrophage phenotypes may have distinct roles in ANCA GN. usCD163 remains an excellent marker to detect active ANCA GN, while ssCD206 seems to be a more prominent marker for risk prediction.

Toll-like receptor signalling in B cells during systemic lupus erythematosus

Simon Fillatreau et al.

Summary: Loss of B cell tolerance to autoantigens in systemic lupus erythematosus (SLE) is driven by TLR7, whereas TLR9 appears to protect against SLE by limiting the stimulatory activity of TLR7. The unique features of Toll-like receptor signaling in B cells implicate it as a therapeutic target in SLE.

Summary: In this study, the C5a receptor inhibitor avacopan was found to be noninferior to prednisone taper in achieving remission at week 26, but superior in achieving sustained remission at week 52 in patients with ANCA-associated vasculitis. All patients received cyclophosphamide or rituximab. The safety and clinical effects of avacopan beyond 52 weeks were not addressed in the trial.

Macrophages in Lung Injury, Repair, and Fibrosis

Peiyong Cheng et al.

Summary: Fibrosis progression in the lung can lead to impaired gas exchange, respiratory failure, or even death. Macrophages play a crucial role in response to lung injury, repair, and fibrosis, with their activation and polarization being key factors in the process. Targeting macrophages may offer potential benefits in alleviating pulmonary fibrosis.

Comparative Histological Subtyping of Immune Cell Infiltrates in MPO-ANCA and PR3-ANCA Glomerulonephritis

Samy Hakroush et al.

Summary: This study analyzed kidney biopsies of patients with ANCA GN and found that different types of immune cell infiltrates were associated with clinicopathological findings, particularly neutrophilic infiltrates were linked to glomerular necrosis and severe kidney injury, while eosinophilic infiltrates were associated with kidney injury and inflammation independent of glomerular lesions, suggesting distinct roles in inflammation and damage in ANCA GN. Plasma cell infiltrates and mononuclear cell infiltrates were also correlated with specific histopathological lesions and disease outcomes. The study suggests that different pathogenic mechanisms may be involved in MPO-ANCA versus PR3-ANCA GN, and may lead to potential new therapeutic targets for specific ANCA subtypes.
