18F-FDG PET Visualizes Systemic STING Agonist-Induced Lymphocyte Activation in Preclinical Models

Thuc M. Le et al.

Summary: This study reveals that systemic STING activation induces metabolic alterations in immune cells, which can be visualized using PET imaging. Using mouse models, the researchers found that STING agonist treatment delays tumor growth and increases immune cell activation. These findings provide a new approach and guidance for pharmacodynamic research on systemic STING agonists.

Immuno-PET Monitoring of CD8+ T Cell Infiltration Post ICOS Agonist Antibody Treatment Alone and in Combination with PD-1 Blocking Antibody Using a 89Zr Anti-CD8+ Mouse Minibody in EMT6 Syngeneic Tumor Mouse

Hasan Alsaid et al.

Summary: The study utilized immuno-PET imaging to investigate the dynamics of CD8(+) T cell infiltration in tumors following ICOS agonist and PD-1 blocking antibody therapy. The results demonstrated the enhanced spatial and temporal resolution of CD8(+) T cell infiltration across tumors and lymphoid tissues using this technique.

CD8-Targeted PET Imaging of Tumor-Infiltrating T Cells in Patients with Cancer: A Phase I First-in-Humans Study of 89Zr-Df-IAB22M2C, a Radiolabeled Anti-CD8 Minibody

Michael D. Farwell et al.

Summary: This study demonstrates that CD8 PET imaging with Zr-89-Df-IAB22M2C is a safe and effective method for visualizing the whole-body biodistribution of CD8+ leukocytes in tumors and reference tissues, and may predict early response to immunotherapy.

A bioluminescence reporter mouse model for visualizing and quantifying CD8+T cells in vivo

Kimberly Bettano et al.

Summary: This study describes a genetically engineered mouse model that can label CD8+ T cells and monitor them longitudinally using in vivo imaging. The specificity of the luciferase signal in CD8+ T cells was confirmed through mRNA correlation analysis. Significant increases in CD8+ T cells, correlated with tumor growth inhibition, were observed in the mouse model. This novel reporter mouse model is an important tool for studying and developing anti-cancer drugs.

Dynamic profiling of immune microenvironment during pancreatic cancer development suggests early intervention and combination strategy of immunotherapy

Jiaqi Yang et al.

Summary: This study reveals two different stages of immunosuppression in the development of pancreatic ductal adenocarcinoma (PDAC), with early stage characterized by abundant Tregs and late stage featuring myeloid suppressive cells. Depletion of PD-L1 and targeting Arg1(+) macrophages effectively triggered anti-tumor immunity, prolonged survival, and generated PDAC-specific immune memory.

In vivo bioluminescence imaging of granzyme B activity in tumor response to cancer immunotherapy

Min Chen et al.

Summary: This study presents a bioluminescence probe, GBLI-2, for real-time imaging of granzyme B activity in tumors undergoing immune checkpoint inhibitor therapy. The GBLI-2 signal correlated with changes in the population of PD-1 and granzyme B-expressing CD8+ T cells in tumors.

Summary: In this study, a transgenic mouse model was generated via CRISPR-mediated genome editing for non-invasive imaging of CD8(+) T cells, providing a convenient and robust tool for understanding fundamental CD8(+) T cell biology. This novel approach offers the potential for experimental applications and preclinical translational studies.
