The cGAS/STING signaling pathway: a cross-talk of infection, senescence and tumors

Peng-Hui Zeng et al.

Summary: The cGAS/STING signaling pathway is an important part of the cytoplasmic DNA sensor that triggers an immune response against microbial infection. However, continuous inhibition of this pathway by viral infection may contribute to tumorigenesis. Additionally, while the cGAS/STING pathway plays a role in anti-tumor immunity and cell senescence, its inflammatory response can also promote tumorigenesis in different contexts. This article discusses the role of cGAS/STING in infection, senescence, and tumors, particularly in terms of progression, to enhance understanding of its mechanism.

How Different Pathologies Are Affected by IFIT Expression

Justin H. H. Franco et al.

Summary: The type-I interferon (IFN) system is the first line of defense against viral pathogens. Secreted IFNs stimulate nearby cells to produce proinflammatory cytokines and antiviral factors. IFIT proteins, known for their antiviral activity, have also been studied in nonviral pathologies.

Personalized RNA neoantigen vaccines stimulate T cells in pancreatic cancer

Luis A. Rojas et al.

Summary: Pancreatic ductal adenocarcinoma (PDAC) is a lethal disease with a high mortality rate, but it can be targeted using mutation-derived T cell neoantigens as vaccines. In this phase I trial, researchers synthesized individualized mRNA neoantigen vaccines from PDAC tumors and administered them to patients along with atezolizumab, autogene cevumeran, and mFOLFIRINOX chemotherapy. The results showed that the autogene cevumeran vaccine induced neoantigen-specific T cells in patients, and those with vaccine-expanded T cells had a longer recurrence-free survival.

Summary: Nucleotide-binding and leucine-rich repeat (NLR) proteins are critical intracellular immune receptors in both animals and plants. This article reviews the latest advances in understanding the structural aspects of NLR inflammasomes and resistosomes, including their activation, assembly, and distinct mechanisms of action. Commonalities and differences between NLR inflammasomes and resistosomes are also discussed.

The chromatin remodeling protein ATRX positively regulates IRF3-dependent type I interferon production and interferon-induced gene expression

Anne-Charlotte Stilp et al.

Summary: In this study, it was discovered that the chromatin remodeling factor ATRX has a co-activating role in innate immunity, regulating chromatin accessibility and transcription of interferons and interferon-stimulated genes. It affects the antiviral immune response by decreasing transcription and secretion of type I interferons, leading to reduced expression of interferon-stimulated genes.

mRNA vaccine for cancer immunotherapy

Lei Miao et al.

Summary: mRNA vaccines have emerged as a promising platform for cancer immunotherapy, showcasing high potency, safe administration, rapid development potentials, and cost-effective manufacturing. However, challenges such as stability, innate immunogenicity, and inefficient in vivo delivery restrict their applications. In addressing these challenges, structural modifications and formulation methods are being researched to enhance the anti-tumor immune response.

STING enhances cell death through regulation of reactive oxygen species and DNA damage

Thomas J. Hayman et al.

Summary: STING functions as an intrinsic regulator of tumor cell survival by controlling ROS homeostasis and affecting DNA damage and therapeutic resistance. Low STING expression is associated with worse tumor outcomes. Pharmacologic activation of STING enhances the effects of ionizing radiation, providing a rationale for therapeutic combinations of STING agonists and DNA-damaging agents.

Cellular networks controlling T cell persistence in adoptive cell therapy

Jack D. Chan et al.

Summary: The differentiation status of tumor-specific T cells greatly impacts their anti-tumor activity, with less differentiated T cells showing better therapeutic effects due to their enhanced expansion and long-term persistence. Adoptive transfer of T cells with stem-like memory or precursor phenotype is associated with improved therapeutic outcomes in cancer patients. Advances in understanding T cell differentiation at the epigenetic and transcriptional levels have led to the development of novel methods for generating more persistent and functional tumor-specific T cells.
