Traditional Chinese medicine technique Governor Vessel electro-acupuncture (GV-EA) has been found to improve the microenvironment of injured spinal cord, and adult stem cell transplantation has potential for reconnecting severed spinal cord tissue and restoring motor and sensory functions. However, no single treatment approach has proven satisfactory, and combining different treatments may be more promising.

Summary: Transplantation of olfactory ensheathing cells (OECs) and their exosomes have shown therapeutic effects for spinal cord injury (SCI) by modulating neuroinflammation, supporting neuronal survival, and promoting angiogenesis. OECs-derived exosomes (OECs-Exo) can be engulfed by microglia, inhibit pro-inflammatory polarization, and promote the numbers of anti-inflammatory cells after SCI. In vitro studies also demonstrate that OECs-Exo can alleviate neuronal death induced by pro-inflammatory microglia, promote neuronal survival and axon preservation, and facilitate functional recovery after SCI.

The Role of TDP-43 in Neurodegenerative Disease

Summary: In recent years, it has been discovered that more and more neurodegenerative diseases exhibit abnormal expression of the TDP-43 protein, including depletion of TDP-43 in the nucleus and accumulation of TDP-43 in the cytoplasm. This TDP-43 proteinopathy suggests the important role of TDP-43 in the pathogenesis of these diseases, and further research is needed to understand the complex pathophysiological mechanisms of TDP-43 in neurodegenerative diseases.

OECs Prevented Neuronal Cells from Apoptosis Partially Through Exosome-derived BDNF

Summary: This study found that exosomes released by olfactory ensheathing cells (OECs) have a protective effect on neuronal survival by modulating the phenotype of macrophages/microglia. Brain-derived neurotrophic factor (BDNF) was identified in these exosomes, and its activation of the TrkB receptor in neurons was demonstrated. Furthermore, it was shown that exosome-derived BDNF partially prevented TNF-alpha-induced neuronal apoptosis.

Summary: This study proposes a method of encapsulating stem cell spheroids in microcapsules using a metal-phenolic network nanofilm, in order to maintain their stemness in an inflammatory microenvironment. The biodegradable MPN biointerface formed acts as a protective barrier for the cells, and exhibits antioxidative, antibacterial, and anti-inflammatory activities, effectively remodeling the inflammatory microenvironment and maintaining stemness.

Summary: Schwann cells in the peripheral nervous system have the ability to promote repair of various tissues, including nerve gaps, skin wounds, digit tips, and tooth regeneration. They are highly motile, secrete signaling molecules, attract macrophages, support neuronal survival, promote axonal regrowth, and interact with other cell types in the regeneration process. The migration of Schwann cells is essential for successful tissue repair, especially in cases of peripheral nerve transection injuries where the cells form cords to guide axon regeneration. Understanding the molecular mechanisms of Schwann cell migration may lead to new therapeutic strategies for peripheral nerve repair.

Peripheral Nerve Regeneration Using a Nerve Conduit with Olfactory Ensheathing Cells in a Rat Model

Summary: The study evaluated the potential of olfactory ensheathing cells (OECs) embedded in a nerve conduit for peripheral nerve injury treatment, showing that a PCL conduit with OECs enhances nerve regeneration and offers a good alternative to autologous nerve grafts. Histomorphometric analysis revealed nerve regeneration and angiogenesis in all groups, with significant differences in the nerve regeneration rate of the muscles between the different treatment groups. No significant differences were observed in nerve conduction velocity, wet muscle weight, and nerve density quantification among the groups.
