Azomethine Ylides-Versatile Synthons for Pyrrolidinyl-Heterocyclic Compounds

Summary: Azomethine ylides are frequently used three-atom components in nitrogen-based [3+2]-cycloaddition reactions with a variety of unsaturated 2 pi-electron components. These reactions demonstrate remarkable regio- and stereoselectivity and have attracted organic chemists' interest in constructing diverse heterocycles with four contiguous stereogenic centers. This review article summarizes the significant [3+2]-cycloaddition reactions of azomethine ylides with various olefinic, unsaturated 2 pi-electron components (acyclic, alicyclic, heterocyclic, and exocyclic ones) reported in the past two decades.

Electrophilicity and nucleophilicity scales at different DFT computational levels

Summary: This study extends the reference electrophilicity and nucleophilicity scales to the most common DFT computational methods used in theoretical organic chemistry studies. Excellent linear correlations were obtained with coefficient of determination R-2 values between 0.97 and 1.00. The study establishes new threshold values for 48 DFT computational levels, enabling direct classification and quantification of the electrophilic and nucleophilic character of molecules without relying on the B3LYP/6-31G(d) scales.

Novel spirooxindole based benzimidazole scaffold: In vitro, nanoformulation and in vivo studies on anticancer and antimetastatic activity of breast adenocarcinoma

The results showed that SP1 exhibited strong inhibitory effects on multiple cancer cell lines and induced apoptosis. Benzodioxole grafted spirooxindole pyrrolidinyl derivatives: synthesis, characterization, molecular docking and anti-diabetic activity

Summary: A highly stereoselective method was developed for synthesizing pyrrolidine and pyrrolizidine containing spirooxindole derivatives. The method showed excellent stereo- and regioselectivity, along with shorter reaction time and high yields. The synthesized compounds exhibited potent inhibition activity against alpha-glucosidase and alpha-amylase enzymes.

Discovery of spirooxindole-ferrocene hybrids as novel MDM2 inhibitors

Summary: A series of spirooxindole-ferrocene hybrids with five or four contiguous chiral centers were designed and synthesized through organocatalysis. Compound 5d was identified as the most potent MDM2 inhibitor, suppressing MDM2-mediated p53 degradation, inducing apoptosis, and promoting oxidative damage. Molecular docking studies have shown that 5d binds to MDM2 by mimicking the Trp23 and Leu26 residues of p53, providing a basis for the development of novel multifunctional MDM2 inhibitors.

The synthesized compounds demonstrated superior cytotoxic activities compared to traditional chemotherapeutic drugs, suggesting their potential as effective anticancer agents. Docking simulations provided insights into the possible binding modes of the compounds within MDM2.
