Immune checkpoint HLA-E:CD94-NKG2A mediates evasion of circulating tumor cells from NK cell surveillance

Summary: Circulating tumor cells (CTCs) shed by primary malignancies are responsible for distant metastasis, but the mechanisms by which CTCs escape immune surveillance are still largely unknown. This study characterizes the transcriptomes of human pancreatic ductal adenocarcinoma CTCs, primary, and metastatic lesions at the single-cell scale. Functional studies reveal that CTCs and natural killer (NK) cells interact through the immune checkpoint molecule pair HLA-E:CD94-NKG2A. Disrupting this interaction enhances NK-mediated tumor cell killing in vitro and prevents tumor metastasis in vivo. Platelet-derived RGS18 plays a role in protecting CTCs from NK-mediated immune surveillance by engaging the HLA-E:CD94-NKG2A immune checkpoint.

A novel whole blood purifier for efficient capture and separation of circulating tumor cells

Summary: Circulating tumor cells (CTCs) play a significant role in predicting the overall survival and progression-free survival of cancer patients. However, current CTCs separation and enrichment techniques have limitations in comprehensively profiling CTC distribution and reducing metastasis. Inspired by hemodialysis, a whole blood purifier (WBP) was designed to capture, enrich and remove CTCs from systemic blood circulation, without causing any organic damages. This approach achieves the high accuracy of liquid biopsy technology and has the potential to be an effective clinical adjuvant therapy for tumor metastasis.

Chimeric nanocomposites for the rapid and simple isolation of urinary extracellular vesicles

Summary: This study developed lactoferrin-conjugated nanoparticles to achieve simple and sensitive isolation of extracellular vesicles (EVs) through electrostatic interaction, physical absorption, and biorecognition. The method showed faster speed, higher efficiency, recovery rate, and purity compared to established methods. The study also confirmed the significantly higher expression of exosomal microRNAs (miRNAs) from cancer cell-derived exosomes. Finally, the method was applied to analyze urinary exosomal miRNAs from prostate cancer patients and healthy controls, revealing significant up-regulation of miR-21 and miR-346 and down-regulation of miR-23a and miR-122-5p in both groups compared to controls. The method provides a rapid, simple, and high-yield approach for human excreta analysis in clinical applications.

Flexible Electronic Catheter Based on Nanofibers for the In Vivo Elimination of Circulating Tumor Cells

Summary: This study reports a flexible electronic catheter that can capture and kill circulating tumor cells (CTCs) with high efficiency. The catheter is assembled from nanofibers with liquid metal-polymer conductor (MPC) electrodes and modified with an epithelial cellular adhesion molecule (EpCAM) antibody to improve specificity. In a rabbit model, the catheter showed 100% killing efficiency for captured CTCs without noticeable damage to normal cells and organs. This electronic catheter offers an alternative strategy for enhancing clinical cancer therapy.
