Sympathetic Innervation Regulates Osteocyte-Mediated Cortical Bone Resorption during Lactation

Summary: Bone is constantly remodeled by the actions of osteoclasts and osteoblasts. However, new research has shown that sympathetic innervation also plays a role in regulating bone resorption by osteocytes. Increased sympathetic activity during lactation stimulates osteocytes to produce netrin-1, which promotes nerve sprouting in the bone. In addition, norepinephrine activates signaling pathways that result in the secretion of bone-degrading enzymes and inhibition of osteoblast differentiation.

Summary: Accumulating evidence suggests that interoception plays a crucial role in maintaining physiological status and metabolic balance by regulating feeding behaviors, glucose balance, and lipid metabolism. Recent research has shown that skeletal metabolism is tightly regulated by the nervous system through skeletal interoception, which contributes to bone homeostasis. This article provides an overview of interoception and its effects on the skeleton and whole-body metabolism, as well as discussing its potential applications in pathological conditions, skeletal pain, and future challenges in understanding and utilizing these processes for therapy.

Summary: In this study, it was found that ascending skeleton interoceptive signaling downregulates hypothalamic NPY expression, inducing lipolysis and promoting osteoblastic bone formation. SMILE plays a key role in this process by inhibiting NPY expression. Inhibition of NPY Y1 receptor can rescue bone loss.
