Neutrophil extracellular traps: from physiology to pathology

Andres Hidalgo et al.

Summary: Neutrophils combat infections and injuries by releasing web-like DNA structures called neutrophil extracellular traps (NETs), which prevent pathogen dissemination or deal with larger microorganisms. However, uncontrolled release of NETs can lead to adverse effects such as vessel occlusion, tissue damage, and prolonged inflammation. This review discusses recent advances in understanding the mechanisms of NET release and functions, as well as the importance of neutrophil heterogeneity in NET formation and composition.

Neutrophils prevent rectal bleeding in ulcerative colitis by peptidyl-arginine deiminase-4-dependent immunothrombosis

Moritz Leppkes et al.

Summary: Bleeding ulcers and erosions are common in active ulcerative colitis, with fibrin layers correlated with rectal bleeding. Neutrophils induce secondary immunothrombosis through PAD4-dependent mechanisms, protecting against acute colitis and rectal bleeding. Deficiency in immunothrombosis can lead to exacerbated colitis and increased bleeding.

Cardiovascular Diseases: Consider Netosis

Chiara Mozzini et al.

Summary: Neutrophil extracellular traps (NETs) are net-like chromatin fibers that are released from dying neutrophils during infections. They play a role in trapping and killing pathogens, but recent studies also suggest their involvement in diseases such as venous thromboembolism, atherosclerosis, myocardial infarction, and heart failure. This review provides evidence of NETs' role in these diseases and discusses the prognostic significance of NETs-related markers in cardiovascular diseases.

Antibiotic use differentially affects the risk of anti-drug antibody formation during anti-TNFα therapy in inflammatory bowel disease patients: a report from the epi-IIRN

Yuri Gorelik et al.

Summary: This study found that antibiotic treatments can affect the risk of developing anti-drug antibodies (ADA) to anti-TNF therapy in patients with inflammatory bowel disease. The use of cephalosporins and penicillins with beta-lactamase inhibitors increased the risk of ADA development, while macrolides and fluoroquinolones reduced the risk.

Impact of Neutrophil Extracellular Traps on Thrombosis Formation: New Findings and Future Perspective

Yilu Zhou et al.

Summary: Thrombotic diseases are a serious threat to human health, and neutrophils and neutrophil extracellular traps (NETs) play a significant role in abnormal thrombus formation. NETs can prevent the spread of pathogens and effectively eliminate them, but they can also contribute to the formation of pathological thrombi.

Summary: This review comprehensively examines the changes in NETs biomarkers associated with thrombosis and their role in thrombus formation. Studies have shown significant alterations in levels of NETs biomarkers in thrombotic patients, highlighting their importance in the development and prognostic assessment of thrombotic diseases. The paper also explores the potential therapeutic implications of NETs and discusses limitations of existing biomarker detection methods.

Berberine treats atherosclerosis via a vitamine-like effect down-regulating Choline-TMA-TMAO production pathway in gut microbiota

Shu-Rong Ma et al.

Summary: This study found that berberine can treat atherosclerosis by reducing trimethylamine-N-oxide (TMAO) production in the gut microbiota. The results showed that berberine interacts with enzymes in the gut microbiota to lower TMAO biosynthesis, resulting in decreased TMAO levels in the blood and interrupted plaque formation in blood vessels. Clinical trials also demonstrated that oral berberine can decrease plaque score and reduce TMA and TMAO levels in atherosclerotic patients.

Extracellular vesicles derived from inflamed murine colorectal tissue induce fibroblast proliferation via epidermal growth factor receptor

Summary: Inflammatory bowel diseases lead to upregulation of proteins in extracellular vesicles, particularly an increase in expression of epidermal growth factor receptor which promotes cell proliferation. The findings suggest that inflamed colon-derived extracellular vesicles promote tumor development by activating fibroblasts.

Summary: Recent studies have shed new light on the biogenesis and functions of long non-coding RNAs (lncRNAs), showcasing their diverse roles in gene regulation and signaling pathways, particularly in the contexts of neuronal disorders, immune responses, and cancer. The discovery of unique biogenesis pathways and subcellular localizations of lncRNAs has opened up potential therapeutic avenues for targeting lncRNAs in various biological and pathophysiological conditions.

Neutrophil Phenotypes and Functions in Trauma and Trauma-Related Sepsis

Andrea Janicova et al.

Summary: Physical trauma can lead to mortality, and the early post-traumatic immune responses can have significant impacts on the development of secondary complications. Neutrophils play a critical role in the inflammatory response to traumatic injury, and dysregulation of their functions can contribute to the development of complications. Targeting neutrophils and their functions may be a potential strategy to improve post-traumatic treatment outcomes.

Exosomes derived from osteogenic tumor activate osteoclast differentiation and concurrently inhibit osteogenesis by transferring COL1A1-targeting miRNA-92a-1-5p

Lijuan Yu et al.

Summary: This study revealed the critical role of PCa exosomes in regulating bone homeostasis and promoting tumor growth in bone. Specifically, miRNAs delivered by PCa exosomes, such as miR-92a-1-5p, were found to play key roles in downregulating type I collagen expression, promoting osteoclast differentiation, and inhibiting osteoblastogenesis. These findings provide new insights into tumor bone metastasis and suggest potential therapeutic targets for PCa bone metastasis.
