Hard-Shelled Glycol Chitosan Nanoparticles for Dual MRI/US Detection of Drug Delivery/Release: A Proof-of-Concept Study

Plasminogen activator-coated nanobubbles targeting cell- bound & beta; 2-glycoprotein I as a novel thrombus-specific thrombolytic strategy

Summary: A new targeted thrombolytic agent consisting of nanobubbles coated with recombinant tissue plasminogen activator (rtPA) and a recombinant antibody specific for cell-bound 132-GPI was developed. The targeted nanobubbles exhibited greater lysing capabilities for platelet-rich blood clots and induced rapid and persistent recanalization of occluded vessels in animal models. This study suggests that targeting cell-bound 132-GPI may be an effective and thrombus-specific thrombolytic strategy.

Dual-drug loaded ultrasound-responsive nanodroplets for on-demand combination chemotherapy

Summary: Phase-changing nanodroplets are nanometric sized constructs that can be vaporized via external stimuli to generate visible bubbles in ultrasound. Our study develops a perfluoropentane core nanodroplet that can load and release paclitaxel and doxorubicin in response to an acoustic trigger. We investigate their loading, release, and biological effects on a breast cancer mouse model, showing enhanced drug-delivery effect and delayed tumor growth rate. Overall, phase-changing nanodroplets are a useful platform for on-demand delivery of drug combinations.

Ultrasound-Responsive Nrf2-Targeting siRNA-Loaded Nanobubbles for Enhancing the Treatment of Melanoma

Summary: siRNA-mediated inhibition of Nrf2 can overcome chemoresistance in malignant tumors. This study designed chitosan-shelled nanobubbles for delivering siRNA against Nrf2, with a new preparation method based on a water-oil-water double-emulsion. The results showed that nanobubbles have promise as a responsive tool for siRNA delivery, able to overcome chemoresistance in melanoma cancer cells.

Exploring chitosan-shelled nanobubbles to improve HER2+immunotherapy via dendritic cell targeting

Summary: This study aims to design a novel immunotherapy nanoplatform for the treatment of HER2 + breast cancer. Specially tailored chitosan-shelled nanobubbles have been developed for the delivery of a DNA vaccine, which are capable of targeting dendritic cells (DCs) and inducing DC activation. In a mouse model, the nanobubbles elicited a specific immune response that was able to slow tumor growth.
