Engineered Salmonella inhibits GPX4 expression and induces ferroptosis to suppress glioma growth in vitro and in vivo

Summary: This study aimed to investigate the therapeutic effects of Salmonella YB1 on glioma growth by inducing ferroptosis. Through mRNA sequencing and microscopy observation, it was found that Salmonella infection altered ferroptosis-related gene expression in U87 glioma cells, leading to reduced expression of GPX4 and increased levels of reactive oxygen species and malondialdehyde. In vivo experiments in mouse tumor xenograft models demonstrated that Salmonella significantly inhibited tumor growth, which could be reversed by Fer-1 treatment. These findings suggest that regulating ferroptosis may improve the efficacy of bacterial cancer therapy.

Selenoprotein GPX1 is a prognostic and chemotherapy-related biomarker for brain lower grade glioma

Summary: The study investigates the association between GPX1 expression and tumor therapy, as well as the prognostic value of GPX1 in various malignancies. The expression of GPX1 was found to be highly correlated with poor prognosis in LGG patients. GPX1 was also associated with tumor purity, tumor mutation burden (TMB), microsatellite instability (MSI), and DNA mismatch repair genes (MMRs). In addition, GPX1 was involved in inflammatory response and apoptosis signaling pathways. The interaction between GPX1 and chemoradiotherapy showed a beneficial clinical effect in LGG patients. GPX1 expression can predict TMZ sensitivity in LGG, providing potential evidence for chemotherapy. Overall, this study provides important insights into the role of GPX1 in human tumors, particularly in LGG.
