Functional Variation in the FAAH Gene Is Directly Associated with Subjective Well-Being and Indirectly Associated with Problematic Alcohol Use

Synaptoproteomic Analysis of the Prefrontal Cortex Reveals Spatio-Temporal Changes in SYNGAP1 Following Cannabinoid Exposure in Rat Adolescence

Summary: The regular use of cannabis during adolescence has been associated with negative life outcomes, but the exact molecular mechanisms are not well understood. This study found that exposure to a synthetic cannabinoid in adolescent rats led to changes in the expression of proteins related to the gamma-aminobutyric acid (GABA)-ergic neurotransmitter system in the prefrontal cortex (PFC). The synaptic protein SYNGAP1 was upregulated in the PFC following prolonged drug abstinence, and this change was specific to adolescent exposure and not seen in adulthood. Similar changes in SYNGAP1 levels were also observed in rats exposed to a natural cannabinoid. The findings suggest a link between cannabinoid exposure and SYNGAP1 changes that are spatio-temporal and developmental in nature.

Fatty acid amide hydrolase inhibition alleviates anxiety-like symptoms in a rat model used to study post-traumatic stress disorder

Summary: In this study, the effects of two novel brain-permeable FAAH inhibitors, ARN14633 and ARN14280, were investigated in a rat model of predator stress-induced long-term anxiety. The results showed that these inhibitors effectively reduced anxiety-like behaviors and were correlated with the inhibition of FAAH activity and increase in FAAH substrate levels. This study supports the potential clinical application of FAAH inhibitors in the management of PTSD.

Association Between Fatty Acid Amide Hydrolase and Alcohol Response Phenotypes: A Positron Emission Tomography Imaging Study With [C-11]CURB in Heavy-Drinking Youth

Phenotypic and genetic analysis of a wellbeing factor score in the UK Biobank and the impact of childhood maltreatment and psychiatric illness

Summary: Wellbeing is an important aspect of mental health that has a moderate level of heritability. This study used data from the UK Biobank resource to investigate the genetic basis of wellbeing and its relationships with mental health and childhood maltreatment. By analyzing the derived wellbeing index, the researchers identified five significant genetic loci associated with wellbeing. They found that the wellbeing index had a genetic correlation with its subcomponents that was stronger than its correlation with mental health phenotypes. The study also revealed a negative association between wellbeing and psychiatric disorders, as well as a moderate genetic correlation between childhood maltreatment and wellbeing.

Genetic diversity fuels gene discovery for tobacco and alcohol use

Summary: This study utilized global genetic diversity data to discover and fine-map genomic loci associated with tobacco and alcohol use. Ancestry-aware transcriptome-wide association studies were conducted to understand the function of these loci. The results demonstrated that increasing sample size and genetic diversity improved the identification and fine-mapping of loci, but polygenic risk scores performed poorly across different ancestries.
