Genome-wide association study of thoracic aortic aneurysm and dissection in the Million Veteran Program

Derek Klarin et al.

Summary: We conducted a GWAS of TAAD and identified 21 risk loci, providing evidence that TAAD is different from other vascular diseases. Our findings suggest that TAAD is not solely inherited through large effect size protein-altering variants and has a genetic architecture similar to other complex traits.

High heritability of ascending aortic diameter and trans-ancestry prediction of thoracic aortic disease

Catherine Tcheandjieu et al.

Summary: This study used automated extraction of ascending aortic diameter from cardiac magnetic resonance images and conducted a genome-wide association study. The polygenic score constructed from multiple loci was able to predict the risk of thoracic aortic aneurysm in different populations. Additionally, the study revealed the causal role of blood pressure in reducing aortic dilation.

Does autosomal dominant polycystic kidney disease increase the risk of aortic aneurysm or dissection: a point of view based on a systematic review and meta-analysis

Raquel Nunes et al.

Summary: This study conducted a systematic review and meta-analysis to analyze the risk of aortic aneurysms and aortic dissections in patients with autosomal dominant polycystic kidney disease (ADPKD). The findings suggested that ADPKD was associated with a significantly higher risk of developing AA/AD.

Double-Hit Mutations in Bicuspid Aortic Valve and Blunt Traumatic Acute Aortic Dissection

Kushtrim Disha et al.

Summary: A young patient underwent surgery for type A acute aortic dissection due to blunt thoracic trauma with a bicuspid aortic valve. Genetic analysis revealed mutations in the NOTCH1 and ACTA2 genes in the patient and his father. Screening of his family members did not show the same mutations.

Novel LOX Variants in Five Families with Aortic/Arterial Aneurysm and Dissection with Variable Connective Tissue Findings

Ilse Van Gucht et al.

Summary: This study found that patients with LOX gene variants have a spectrum of aortic and arterial aneurysmal diseases, often combined with connective tissue abnormalities. Some patients developed TAAD in early life, while others had normal aortic diameters in advanced age.

Cardiac Involvement in Autosomal Dominant Polycystic Kidney Disease

Letizia Spinelli et al.

Summary: Cardiovascular disorders are the main complication in autosomal dominant polycystic kidney disease (ADPKD), with dysfunction of the polycystin proteins in the cardiovascular system playing a contributing factor to these abnormalities. Specifically, the effects of polycystin proteins' deficiency on cardiomyocyte function have been considered in studies.
