Vitamin D3 and COVID-19 Outcomes: An Umbrella Review of Systematic Reviews and Meta-Analyses

Summary: This study found that vitamin D metabolism and genetic polymorphisms of VDR are associated with the development of CD. Healthy controls have significantly higher levels of 25(OH)D serum than CD patients, and the T-allele of Fok1 is a predisposing allele for CD. The results of this study suggest that reduced serum levels of 25(OH)D and the Fok1 T-allele of VDR play a critical role in immunomodulation and maintaining barrier integrity, which are majorly implicated in CD.

The ability of P450s to catalyze various reactions makes them promising enzymes for numerous applications, and future research in this field is encouraged.

Gene Polymorphisms and Biological Effects of Vitamin D Receptor on Nonalcoholic Fatty Liver Disease Development and Progression

Summary: Nonalcoholic fatty liver disease (NAFLD) is a common chronic liver disease with a rising global prevalence. The vitamin D/vitamin D receptor (VDR) axis plays a significant role in the development and progression of NAFLD. Gene polymorphisms affecting the regulation of the VDR gene are associated with the presence and severity of NAFLD, as they influence adipose tissue activity, fibrosis, and hepatocellular carcinoma (HCC) development. Vitamin D binds to the hepatic VDR, activating transcriptional activity and regulating gene expression related to inflammation and fibrosis, as well as inducing intracellular signal transduction through VDR-mediated activation of Ca2+ channels. Elucidating the genetic and molecular background of VDR in NAFLD pathophysiology can lead to the development of VDR agonists as potential therapeutic targets.

In men, sufficient vitamin D levels and moderate physical exercise were linked to decreased risk of very low NK cell activity, while physical exercise significantly reduced the risk in individuals over 60 years old, regardless of gender.

Summary: This ancillary analysis found that a single high dose of 200,000 IU of vitamin D-3 did not significantly reduce the length of hospital stay for patients with severe 25-hydroxyvitamin D deficiency and COVID-19. The study also showed that vitamin D-3 significantly increased serum 25-hydroxyvitamin D levels, but did not impact other clinical outcomes.
