Summary: In this study, we investigated the regulatory mechanism of long non-coding RNA H19 (H19) in BMP9-induced angiogenesis. We demonstrated that H19 was highly expressed in mesenchymal stem cells (MSCs), and silencing H19 attenuated BMP9-induced osteogenesis and angiogenesis in MSCs. Furthermore, down-regulation of H19 inhibited BMP9-mediated blood vessel formation and subsequent bone formation. Mechanistically, H19 promoted the phosphorylation of p53 by direct interaction and phosphorylation binding, thereby enhancing Notch1 expression and activation of Notch1-targeting genes.
