Upregulation of LRRC8A by m(5)C modification-mediated mRNA stability suppresses apoptosis and facilitates tumorigenesis in cervical cancer

Summary: LRRC8A is upregulated in cervical cancer and associated with poor prognosis. It maintains cell survivals under hypotonic condition and promotes tumorigenesis through apoptosis suppression. NSUN2-mediated m(5)C modification upregulates LRRC8A expression, and loss of NSUN2 suppresses the proliferation and metastasis of cervical cancer cells. The NSUN2-m(5)C-LRRC8A axis is crucial and could be a potential therapeutic target for cervical cancer.

Structure of a volume-regulated heteromeric LRRC8A/C channel

Summary: Volume-regulated anion channels (VRACs) play a role in cellular response to osmotic swelling. These membrane proteins are composed of heteromeric assemblies of LRRC8 subunits, with compositions affecting permeation properties. In this study, the structure of murine LRRC8A/C channels was characterized. The assembly showed a predominant organization with an A:C ratio of two, with LRRC8A subunits stabilizing a closed state of the channel while LRRC8C subunits enhancing the activation properties.

LRRC8A is responsible for exosome biogenesis and volume regulation in colon cancer cells

Summary: LRRC8A is a component of exosomes released by colon cancer HCT116 cells and its downregulation inhibits cell growth and induces apoptosis. Chloride channel blockers DCPIB and NPPB also inhibit cell growth and induce apoptosis without altering exosome quantity and proportion. Overexpression of LRRC8A leads to the release of distinct populations of exosomes and eliminates changes in exosome ratios during hypotonic challenge.

Summary: Volume-regulated anion channels (VRACs) mediate volume regulation and solute efflux from cells, but their composition and structure remain unclear. In this study, the authors developed novel homomeric LRRC8 chimeric channels that mimic native VRACs. They found that the LRRC8C-LRRC8A(IL1(25)) chimera has a seven-subunit structure, a large pore size, and normal pharmacology compared to non-functional homomeric channels. Lipid-like densities were also observed in the channel, suggesting a role in gating and regulation.

ATP-releasing SWELL1 channel in spinal microglia contributes to neuropathic pain

Summary: Following peripheral nerve injury, activation of purinergic signaling, mediated by extracellular ATP release, is essential for microglia activation and neuropathic pain. However, the mechanisms of ATP release are not well-understood. This study identifies the volume-regulated anion channel (VRAC) as an ATP-releasing channel activated by the inflammatory mediator sphingosine-1-phosphate (S1P) in microglia. Knockout mice lacking the VRAC subunit Swell1 show reduced ATP release, decreased microgliosis, neuronal hyperactivity, and attenuated neuropathic pain behaviors. Additionally, a high-throughput screen identifies dicumarol as a novel and potent VRAC inhibitor, which can alleviate mechanical allodynia in nerve-injured mice. These findings establish ATP-releasing VRAC in microglia as a crucial determinant of neuropathic pain and a potential therapeutic target.

Recent progress has been made in developing the molecular pharmacology of VRAC through screening FDA-approved drugs, leading to the discovery of novel VRAC inhibitors and activators. This sets the stage for further research on the molecular pharmacology and therapeutic potential of VRAC.
