Integrated Machine Learning and Bioinformatic Analyses Constructed a Novel Stemness-Related Classifier to Predict Prognosis and Immunotherapy Responses for Hepatocellular Carcinoma Patients

Dongjie Chen et al.

Summary: This study proposes a novel classification system for hepatocellular carcinoma (HCC) based on cancer stem cells (CSCs) that can predict patients' response to immunotherapy. Using RNA-seq datasets and machine learning algorithms, the study computes a stemness index (mRNAsi) and performs clustering analysis to categorize HCC patients into different stemness subtypes. This stemness-based classification system can facilitate prognostic prediction and guide clinical strategies for immunotherapy and targeted therapy in HCC.

Molecular Characteristics, Clinical Significance, and Cancer Immune Interactions of Angiogenesis-Associated Genes in Gastric Cancer

Xin Qing et al.

Summary: This study systematically assessed the association between angiogenesis and gastric cancer (GC). The authors identified two molecular subtypes and determined the correlation between angiogenesis-associated genes mutations, clinicopathological characteristics, prognosis, immune cell infiltration, and tumor microenvironment. They established an AAG_score for predicting overall survival (OS) and created a reliable nomogram for clinical viability. The study provides insights into the characteristics of TME and potential immunotherapy strategies in GC.

Programmed Death-Ligand 1 (PD-L1) as Immunotherapy Biomarker in Breast Cancer

Martin Nunez Abad et al.

Summary: This article examines the role of PD-L1 as a biomarker in different subtypes of breast cancer, including clinical trials with immune checkpoint inhibitors and their results. PD-L1 appears to be associated with worse prognosis in the hormone receptor positive subtype. In triple-negative breast cancer patients, immune checkpoint inhibitors have shown good treatment response, especially when combined with chemotherapy. However, the utility of PD-L1 as a predictive biomarker in other breast cancer subtypes remains unclear.

Integrated analysis of single-cell and bulk RNA sequencing data reveals a pan-cancer stemness signature predicting immunotherapy response

Zhen Zhang et al.

Summary: By analyzing single-cell RNA sequencing data from ICI-treated patients, it was found that cancer stemness may be associated with resistance to ICI treatment. The developed novel stemness signature (Stem.Sig) showed improved predictive performance for ICI response across multiple cancers, potentially serving as a competitive tool for patient selection in immunotherapy.

Liver glycogen phosphorylase is upregulated in glioblastoma and provides a metabolic vulnerability to high dose radiation

Christos E. Zois et al.

Summary: This study reveals the significant role of glycogen metabolism in glioblastoma (GBM) cancer. Inhibition of glycogen degradation sensitizes GBM cells to high-dose ionising radiation (IR), suggesting that PYGL may serve as a potential novel target for GBM treatment.

Summary: In this study, GSCALite was upgraded to GSCA, a gene set-based cancer analysis platform that integrates multi-omics data from four public sources and provides functionalities for exploring, analyzing, and visualizing gene expression, genomic variations, tumor immune infiltration, drug sensitivity, and their associations with clinical outcomes. Case studies proved the advantage of GSCA over single gene analysis in discovering novel associations between gene set expression/mutation and clinical outcomes in different cancer types.

Immune suppression in gliomas

Matthew M. Grabowski et al.

Summary: Gliomas evade immune detection and destruction through mechanisms such as dysfunctional T cells and NK cells, expansion of regulatory T cells and myeloid-derived suppressor cells, M2 phenotypic transformation in macrophages/microglia, upregulation of immunosuppressive factors, tumor microenvironment hypoxia, and iatrogenic effects of immunosuppressive treatments. Future research should focus on addressing these immunosuppressive mechanisms to develop treatment options with improved survival outcomes.

Divya Thomas et al.

Summary: Glycosylation is a crucial post-translational modification that plays a significant role in health and diseases. Aberrantly glycosylated proteins have been recognized as modulators of cancer cell phenotype, highlighting the potential of glycans as novel clinical biomarkers. The roles of glycans in tumor progression emphasize their promising target for cancer therapy.

Identification of diverse tumor endothelial cell populations in malignant glioma

Jeff C. Carlson et al.

Summary: This study established molecular and morphological benchmarks for tumor associated vessels and tumor derived endothelial cells during glioblastoma progression. The research found progressive alteration of vessel function and morphogenesis over time, with TDECs identified as a rare subpopulation contributing to vessels within the tumor. Additionally, transcriptional profiling demonstrated molecular distinctiveness and extensive heterogeneity within both TAVs and TDECs, with these findings also present in human glioma.
