Senescent Cells: A Therapeutic Target in Cardiovascular Diseases

Masayoshi Suda et al.

Summary: Senescent cell accumulation is observed in age-associated diseases including cardiovascular diseases. Senescent cells lack proliferative capacity and secrete senescence-associated secretory phenotype (SASP) factors that may cause or worsen cardiovascular diseases. Therapies targeting senescent cells, especially senolytic drugs, have shown potential in delaying or treating age-associated diseases.

Geroscience: the need to address some issues

Eric Le Bourg

Summary: The geroscience field suggests that delaying the ageing process itself could potentially delay all age-related diseases, instead of individually treating each disease. However, there are some issues that temporarily hinder full adherence to this perspective, such as the risk of over-enthusiasm or scientism, and the assumption that animal model results can easily be applied to humans.

Diabetes-Induced Cellular Senescence and Senescence-Associated Secretory Phenotype Impair Cardiac Regeneration and Function Independently of Age

Fabiola Marino et al.

Summary: Diabetes mellitus affects the biology of cardiac stem/progenitor cells, inhibiting their regenerative potential. By clearing senescent cells, the function of these cells can be restored, resulting in improved cardiac function in diabetic patients.

Pharmacological clearance of senescent cells improves cardiac remodeling and function after myocardial infarction in female aged mice

Nadia Salerno et al.

Summary: Cardiovascular diseases are closely related to aging, and the interaction between sex and age plays an important role in the development and progression of these diseases. Cardiac male aging is characterized by the accumulation of senescent cells, but it is unclear whether this is also a significant feature in the aged female heart, and whether senolytics can promote myocardial repair and regeneration in aged female mice after myocardial infarction. The study found that senescent cells accumulate in the aged female hearts, and removing these cells is a key therapeutic target for efficient repair of the aged female heart.

Metformin in aging and aging-related diseases: clinical applications and relevant mechanisms

Sheng Chen et al.

Summary: Aging is a natural process that plays a critical role in the development of various diseases. Metformin, an antidiabetic drug, has been reported to delay aging and provide protection against aging-related diseases by impacting key events in the aging process.

Senescence in Post-Mitotic Cells: A Driver of Aging?

Thomas von Zglinicki et al.

Summary: Cell senescence is not limited to proliferating cells, but can also occur in non-dividing cells. Different senescence domains interact with each other and may play a significant role in mammalian aging.

Strategies for late phase preclinical and early clinical trials of senolytics

Erin O. Wissler Gerdes et al.

Summary: Accumulation of senescent cells is associated with various age-related and chronic diseases, making them potential targets for intervention. Preclinical and early clinical trials with senolytics have shown promising results, but more clinical trials are needed to further evaluate their efficacy across different aging and disease models.

Senescence and senolytics in cardiovascular disease: Promise and potential pitfalls

W. Andrew Owens et al.

Summary: Ageing is linked to increased risk of cardiovascular diseases, with cell senescence believed to play a role in the development of these diseases. Senolytics targeting aging cells show promise as a potential treatment for cardiovascular diseases, but may also pose complications that need to be considered in the context of the cardiovascular system.

The Emergence of Senescent Surface Biomarkers as Senotherapeutic Targets

Martina Rossi et al.

Summary: Senetherapeutics are emerging to suppress the detrimental effects of senescence by targeting either senomorphics or senolytics. The main goal is to suppress the proinflammatory senescence-associated secretory phenotype and promote immune recognition and elimination of senescent cells. One attractive approach is targeting specific molecules or proteins on the surface of senescent cells for senolysis.
