This study identifies histone deacetylase 4 (HDAC4) as a sleep-regulating molecule and demonstrates that the LKB1-SIK3-HDAC4 signaling cascade plays a role in sleep regulation. Additionally, the study shows that SIK3 signaling in excitatory neurons in the cerebral cortex and hypothalamus positively regulates EEG delta power and non-rapid eye movement sleep (NREMS) duration, respectively. These findings provide insights into the intracellular events and circuit-level mechanisms that control sleep.
